目的 探讨环孢素A(CsA)对小鼠体内CD4+Foxp3+调节性T淋巴细胞的作用及机制.方法 选择雄性C57BL/6小鼠20只,随机平均分为两组.实验组小鼠腹腔注射环孢素A20 mg·kg-1·d-1×2周;对照组小鼠腹腔注射等体积的磷酸盐缓冲液(PBS)×2周.2周后,两组小鼠在无菌条件下经眼球采集外周血,并分离脾脏和胸腺,制备单细胞悬液.采用流式细胞仪检测小鼠外周血、脾细胞和胸腺细胞悬液中的CD4+Foxp3+T淋巴细胞的比例.结果 实验组小鼠外周血、脾细胞和胸腺细胞悬液中CD4+Foxp3+T淋巴细胞的比例分别为(0.581±0.089)%、(2.189±0.046)%和(0.472±0.049)%,对照组的比例分别为(1.751±0.227)%、(3.684±0.169)%和(1.412±0.188)%,两组间比较,差异均有统计学意义(P<0.001).结论 CsA可以损害小鼠体内调节性T淋巴细胞的发育,从而减少调节性T淋巴细胞的数量.

목적 탐토배포소A(CsA)대소서체내CD4+Foxp3+조절성T림파세포적작용급궤제.방법 선택웅성C57BL/6소서20지,수궤평균분위량조.실험조소서복강주사배포소A20 mg·kg-1·d-1×2주;대조조소서복강주사등체적적린산염완충액(PBS)×2주.2주후,량조소서재무균조건하경안구채집외주혈,병분리비장화흉선,제비단세포현액.채용류식세포의검측소서외주혈、비세포화흉선세포현액중적CD4+Foxp3+T림파세포적비례.결과 실험조소서외주혈、비세포화흉선세포현액중CD4+Foxp3+T림파세포적비례분별위(0.581±0.089)%、(2.189±0.046)%화(0.472±0.049)%,대조조적비례분별위(1.751±0.227)%、(3.684±0.169)%화(1.412±0.188)%,량조간비교,차이균유통계학의의(P<0.001).결론 CsA가이손해소서체내조절성T림파세포적발육,종이감소조절성T림파세포적수량.
Objective To investigate the effeet of eyclosporine A (CsA) on CD4+ Foxp3+ T cells in C57BL/6 mice and the underlying mechanism.Methods C57BL/6 mice, aging 8~ 10 weeks, were administered intraperitoneally with CsA at a dose of 20 mg· kg-1·d-1 for 2 weeks, while the control mice given sterile PBS.After 2 weeks, peripheral blood was collected, and splenoeytes and thymoeytes were prepared for the detection of CD4 + Foxp3 + T cells by using flow cytometry.Results The proportion of CD4+ Foxp3+ T cells in peripheral blood, spleen and thymus of CsA-treated mice was (0.581 ± 0.089) %, (2.189 ± 0.046) % and (0.472 ± 0.049) %, while that in PBS-treated mice was (1.751 ± 0.227) %, (3.684 ± 0.169) % and (1.412 ± 0.188) %, respectively (P < 0.001).Conclusion The reduction of CD4 + Foxp3+ T cells in CsA-treated mice suggests that CsA can inhibit the development of regulatory T cells.
