目的 通过研究国人原发性及复发性垂体瘤中AIP基因的突变,分析该基因的突变及与垂体瘤复发的相关性.方法 选取病理证实的国人原发性垂体瘤38例及复发性垂体瘤29例,对所有标本进行DNA提取,然后进行PCR扩增及DNA测序比对,并对该基因的突变与垂体瘤复发的相关性进行统计学分析.结果 原发性垂体瘤中AIP基因突变率为13.2％,而在复发性垂体瘤中AIP基因突变率为40.7％.经过卡方检验,两组样本中的突变率差异有统计学意义(P=0.011,P＜0.05).结论 国人原发性及复发性垂体瘤中AIP基因均出现突变,但后者的突变率更高,可见AIP基因的突变与垂体瘤复发存在相关性,提示对AIP基因的研究可能发现一种能早期诊断及治疗垂体瘤复发的新途径.

목적 통과연구국인원발성급복발성수체류중AIP기인적돌변,분석해기인적돌변급여수체류복발적상관성.방법 선취병리증실적국인원발성수체류38례급복발성수체류29례,대소유표본진행DNA제취,연후진행PCR확증급DNA측서비대,병대해기인적돌변여수체류복발적상관성진행통계학분석.결과 원발성수체류중AIP기인돌변솔위13.2％,이재복발성수체류중AIP기인돌변솔위40.7％.경과잡방검험,량조양본중적돌변솔차이유통계학의의(P=0.011,P＜0.05).결론 국인원발성급복발성수체류중AIP기인균출현돌변,단후자적돌변솔경고,가견AIP기인적돌변여수체류복발존재상관성,제시대AIP기인적연구가능발현일충능조기진단급치료수체류복발적신도경.
Objective To investigate the mutations of AIP gene in primary and recurrent pituitary adenomas in Chinese,and to analyze the correlation between the mutations of AIP gene and the relapse rate of pituitary adenomas.Method 38 primary pituitary adenomas and 29 recurrent pituitary adenomas which were verified by pathology were collected,and all specimens were not accepted concurrent chemoradiotherapy and chemo - treatment.DNA was extracted from the specimens,and then proceeded the PCR reaction,and DNA sequence.Results After comparing the sequence investigated with the sequence in Genebank,the mutation in 5 specimens from 38 primary pituitary adenomas and in 12 specimens from 29 recurrent pituitary adenomas were found.There was a significantly difference between the two specimans (x2 =0.009,P =0.011,P ＜ 0.05).Conclusions There are mutations in both Chinese primary pituitary and recurrent pituitary adenomas,but the later had the higher rate of mutations of AIP gene.Exploring the mutation of AIP gene may be a new method for early diagnosis and therapy of recurrent pituitary adenoma.
