Angiotnsin-Converting Enzyme Inhibitors
目的 观察贝那普利及螺内酯对阿霉素肾病大鼠肾小管间质损伤的改善作用.方法 清洁雄性SD大鼠42只,随机抽取7只为正常对照组(A组),余下分35只造模,6周后29只造模成功的大鼠分为:B组为模型对照组,C组为贝那普利治疗组,D组为螺内酯治疗组,E组为贝那普利和螺内酯联合治疗、组.分别于6,12,18周末收集24小时尿液,并于18周末处死大鼠收取血液标本待测,取出肾组织进行病理组织形态学观察,并采用免疫组织化学方法检测肾小管间质VCAM-1的表达.结果 和模型组比较,螺内酯组及联合治疗组大鼠血钾轻度升高;贝那普利组、螺内酯组及联合治疗组血清VCAM-1及胆固醇、甘油三酯浓度均降低,蛋白尿减少,血清白蛋白浓度升高;两药联用效果更为明显.结论 贝那普利与螺内酯联用可进一步降低大鼠血清VCAM-1浓度,同时减少尿蛋白,但有增高血钾的风险,两药联用对阿霉素肾病大鼠肾小管间质损伤具有更好的保护作用.

목적 관찰패나보리급라내지대아매소신병대서신소관간질손상적개선작용.방법 청길웅성SD대서42지,수궤추취7지위정상대조조(A조),여하분35지조모,6주후29지조모성공적대서분위:B조위모형대조조,C조위패나보리치료조,D조위라내지치료조,E조위패나보리화라내지연합치료、조.분별우6,12,18주말수집24소시뇨액,병우18주말처사대서수취혈액표본대측,취출신조직진행병리조직형태학관찰,병채용면역조직화학방법검측신소관간질VCAM-1적표체.결과 화모형조비교,라내지조급연합치료조대서혈갑경도승고;패나보리조、라내지조급연합치료조혈청VCAM-1급담고순、감유삼지농도균강저,단백뇨감소,혈청백단백농도승고;량약련용효과경위명현.결론 패나보리여라내지련용가진일보강저대서혈청VCAM-1농도,동시감소뇨단백,단유증고혈갑적풍험,량약련용대아매소신병대서신소관간질손상구유경호적보호작용.
Objectives To observe the bettering effect of Benazepril and Spironolactone on renal tubule mesenchyme damage in adriamycin ( ADR)-induced nephrotic rat models. Methods Among 42 cleaned male Sprague-Dawley (SD) rate, 7 were randomly taken as normal control group (A group ). And the rest 35 rats were induced by tail intravenous injection of ADR( 1.75mg/kg) . twenty-nine ADR-induced nephrotic male Sdrate were randomly divided into model control group(B group) , Benazepril treatment group(C group) , Spironolactone treatment group( D group) , and Benazepril-Spironolactone-combined treatment group(E group). At the end of 6,12, and 18 weeks, the samples of 24h urine were collected. And at the end of the 18th week, the rats were executed, and blood specimen was collected to be detected. Nephridial tissue was taken and pathomorphological observation was carrying out. Meanwhile, the expression of renal tubule mesenchyme VCAM-1 was detected using immunohisto chemistry technique. Results Compared with B group, serum potassium of rate in D and E group increased subtlely. In C, D, and E group, the levels of serum VCAM-1 x cholesterol and triglycerides all decreased, the proteinuria also decreased, while the levels of serum albumin increased. And the joint treatment had a more obvious effect.Conclusions Combined using Benazepril and Spironolactone can decrease the levels of serum VCAM-1and urine protein, but it also has the risk of serum potassium increasing. And it has a protecting role for renal tubule mesenchyme damage in adriamycin (ADR)-induced nephrotic rate.
