目的检测和分析人脑胶质瘤中CCND1基因的转录与表达,及其对肿瘤发生、发展的生物学意义.方法采用免疫组化、原位杂交法检测52例人脑胶质瘤及8例正常人脑组织中Cyclin D1 mRNA蛋白以及增殖细胞核抗原(PCNA)的表达水平.结果脑胶质瘤中CD1蛋白及mRNA呈过度表达,其标记指数和阳性率随肿瘤的恶性程度增高而增加.两者的标记指数之间、以及与PCNA标记指数之间均为显著正性相关.结论人脑胶质瘤中CCND1过度转录和表达,随胶质瘤临床病理分级增加而增高,同时与肿瘤细胞增殖状态密切相关,提示CCND1基因的异常转录和表达在胶质瘤的发生与发展中起着重要的促进作用.

Objective To investigate the transcription and expression of CCND1 and its role in oncogenesis and progression of human brain gliomas.Methods The expression of CCND1 mRNA and cyclin D1 protein was detected by in situ hybridization and immunohistochemistry respectively in 8 normal brain cases and 52 glioma cases ranging from WHO grade Ⅰ to Ⅳ. Proliferation activity was determined by proliferating cell nuclear antigen (PCNA) immunohistochemical labeling. Results CCND1 was aberrantly transcripted and expressed in gliomas of each malignancy grade. Mean labeling indices (LI) and positive rates for CD1 protein and mRNA significantly increased with the grade of pathological malignancy. Moreover close positive correlations had been found between LI of CD1 and mRNA and that of PCNA.ConclusionCCND1 is overexpressed in human glioma with a tendency to increase with pathological malignancy. In addition, it is positively associated with the proliferation of glioma cells. This suggests that CCND1 may act as an important promotive factor in the oncogenesis and progression of gliomas.
