目的 探讨长时间应用大剂量的左旋精氨酸对幼鼠学习记忆功能和神经系统可塑性的影响.方法 分别给初断乳的大鼠灌胃大剂量的NO前体左旋精氨酸(L-arginine,L-Arg)[600 mg/(kg·d),L-Arg组]和同样剂量的蒸馏水(对照组),每组各10只,持续3个月.用Morris水迷宫检测大鼠的学习记忆功能,用免疫组化技术和HE染色检测大脑皮质和海马CA1、CA3、DG区的神经元型一氧化氮合酶(nNOS)和c-fos基因的表达以及神经细胞数目的 变化.结果 实验结果显示大鼠寻台潜伏期的时间和nNOS的表达在L-Arg组明显较对照组升高,但c-fos基因的表达和大脑皮质及海马CA1、CA3、DG区的神经元数目在L-Arg组明显较对照组降低(P<0.05或P<0.01).结论 长时间应用大剂量的NO前体L-Arg可抑制幼鼠的学习记忆功能和神经系统的可塑性.

Objective To study the effects of long-lasting administration of large dose of L-arginine(L-Arg) on the ability of learning and memory and the plasticity of nervous system in junior rats. Methods The weanling rats were commensurate distilled water (control group,n=10) for three months,respectively.The ability of learning and memory was observed by Morris water maze.Then nNOS (neuronal NOS) or c-fos expression with immunocytochemisity and the number of neuronal cells with HE staining in cerebral cortex and CA1,CA3,DG of hippocampus were detected. Results The latency to find the platform and the expression of nNOS were significantly higher in L-Arg group than that in control group,but the expression of c-fos and the number of neuronal cells in cerebral cortex and CA1,CA3,DG of hippocampus were markedly lower in L-Arg group than that in control group (P<0.05 or P<0.01). Conclusion The results suggest that long-lasting use of L-Arg with large dose can inhibit the ability of learning and memory and the plasticity of nervous system in the developing brain.
