目的研究大鼠同种异体心脏移植免疫耐受的特异性。方法建立大鼠异位心脏移植模型,应用抗淋巴血清(ALS)及/或胸腺内注射脾细胞抗原预处理受体,21天后进行心脏移植。经过心脏移植并产生免疫耐受的受体鼠在心脏移植60天后再次接受供体鼠的皮肤移植。结果同时应用ALS及胸腺注射脾细胞抗原组的移植心平均存活时间较其它组显著延长(平均>72天)。移植的皮肤在移植心耐受组中平均存活7.4天即被排斥,这与未处理组的皮肤移植无差别,但移植皮肤的排斥并不影响移植的心脏。结论同种异体心脏移植产生的特异性免疫耐受是胸腺内注射供体脾细胞和腹腔注射ALS共同作用的结果。胸腺预注射供体脾细胞并腹腔注射ALS诱导产生的免疫耐受具有器官特异性及组织特异性。

목적연구대서동충이체심장이식면역내수적특이성。방법건립대서이위심장이식모형,응용항림파혈청(ALS)급/혹흉선내주사비세포항원예처리수체,21천후진행심장이식。경과심장이식병산생면역내수적수체서재심장이식60천후재차접수공체서적피부이식。결과동시응용ALS급흉선주사비세포항원조적이식심평균존활시간교기타조현저연장(평균>72천)。이식적피부재이식심내수조중평균존활7.4천즉피배척,저여미처리조적피부이식무차별,단이식피부적배척병불영향이식적심장。결론동충이체심장이식산생적특이성면역내수시흉선내주사공체비세포화복강주사ALS공동작용적결과。흉선예주사공체비세포병복강주사ALS유도산생적면역내수구유기관특이성급조직특이성。
Objective This study investigated the durability and specificity of tolerance produced by pretreating the recipient with antilymphocyte serum (ALS) and donor spleen cells given intrathymically.Methods We investigated the specificity of the tolerance produced by this pretreatment in a rat model of heterotopic heart transplantation with Wistar donors and SD recipients pretreated SD rats received 1 ml ALS intraperitoneally and 2.5×107 Wistar splenocytes intrathymically,followed 21 days later by Wistar cardiac transplantation.After 60 days tolerant SD recipients underwent subsequent transplantation with Wistar skin.Results Intrathymic injection of spleen cells combined with ALS intraperitoneally could significantly prolong graft. The Wistar cardiac allograft survived long term(mean 72 days) inpretreated recipients. Who were given no subsequent immunosuppression.After 60 days,tolerant SD recipients underwent subsequent transplantation with Wistar skin. Skin allograft survival in this group(mean 7.4 days)was not different from that in SD recipient without pretreatment,but rejection of skin grafts had no effect on the heart grafts.Conclusion Our results confirm the tolerance induced in a cardiac allograft by intrathymic and ALS pretreatment is permanent and it is specific for donor strain and tissue type.Intrathymic pretreatment therefore appears to produce a permanent alteration in immune response to donor tissues.
