采用正交试验得出了提取山药糖蛋白的优化试验条件,即提取温度40℃、提取时问2 h、料液比1:50.山药经水浸提分离、浸提液脱蛋白、透析、乙醇沉淀物经DEAE-52及Sephadex G-75柱层析得到两种糖蛋白组分GLP-Ⅰ和GLP-Ⅱ,经SDS-聚丙烯酰胺凝胶电泳,相对分子量分别是13000和38000.

Utilizing orthogonal test of the extraction of glycoprotein from Chinese yam,this article put forward an optimum extracting condition as follows:extracting temperature was 40℃,extracting time was 2 h,material-water ratio was 1:50.Two glycoproteins named GLP-Ⅰ and GLP-Ⅱ were isolated from Chinese yam with water extraction,Sevage deprotein,dialysis,alcohol sedimentation,DEAE-52 and Sephadex G-75 column chromatography.Their average molecular weight were 13000 Da and 38000 Da with SDS-PAGE.
