目的 探讨骨髓增生异常综合征(MDS)患者骨髓克隆细胞表面胰岛素样生长因子Ⅰ型受体(IGF-Ⅰ R)的表达状况.方法 联合应用DNA荧光原位杂交(FISH)、免疫细胞化学(碱性磷酸酶-抗碱性磷酸酶系统,APAAP)技术检测40例MDS患者骨髓克隆细胞的IGF-Ⅰ R表达.结果 MDS患者骨髓克隆细胞表面IGF-Ⅰ R表达明显高于正常细胞[分别为(84.5±14.2)%和(11.4±12.1)%](P<0.01).MDS患者骨髓克隆细胞阳性率与有核细胞IGF-Ⅰ R表达率明显相关(r=0.929,P<0.01).高危组MDS患者骨髓克隆细胞IGF-Ⅰ R表达与低危组差异无统计学意义.不同异常核型的克隆细胞IGF-Ⅰ R表达率差异也无统计学意义.结论 IGF-Ⅰ R表达阳性提示细胞的恶性增殖倾向,可能作为MDS骨髓恶性克隆细胞的标志物.
目的 探討骨髓增生異常綜閤徵(MDS)患者骨髓剋隆細胞錶麵胰島素樣生長因子Ⅰ型受體(IGF-Ⅰ R)的錶達狀況.方法 聯閤應用DNA熒光原位雜交(FISH)、免疫細胞化學(堿性燐痠酶-抗堿性燐痠酶繫統,APAAP)技術檢測40例MDS患者骨髓剋隆細胞的IGF-Ⅰ R錶達.結果 MDS患者骨髓剋隆細胞錶麵IGF-Ⅰ R錶達明顯高于正常細胞[分彆為(84.5±14.2)%和(11.4±12.1)%](P<0.01).MDS患者骨髓剋隆細胞暘性率與有覈細胞IGF-Ⅰ R錶達率明顯相關(r=0.929,P<0.01).高危組MDS患者骨髓剋隆細胞IGF-Ⅰ R錶達與低危組差異無統計學意義.不同異常覈型的剋隆細胞IGF-Ⅰ R錶達率差異也無統計學意義.結論 IGF-Ⅰ R錶達暘性提示細胞的噁性增殖傾嚮,可能作為MDS骨髓噁性剋隆細胞的標誌物.

Objective To probe the expression level of insulin-like growth factor- Ⅰ receptor( IGF-Ⅰ R) in malignant clone cells of myelodysplastic syndromes (MDS).Method The combination of fluorescence in situ hybridization (FISH) and immunochemistry ( alkaline phosphatase anti-alkaline phosphatase,APAAP) was used to detect the expression of IGF- Ⅰ R in the bone marrow cells of 40 MDS patients with abnormal karyotypes.Results The average IGF- Ⅰ R expression level on the clone cells[ (84.5 ± 14.2)% ]from the MDS cases was markedly elevated compared to the corresponding level on normal cells [ ( 11.4 ±12.1 )% ].And the percentages of malignant clone cells in all 40 MDS cases were significantly correlated with the relevant percentages of IGF- Ⅰ R positive nucleated cells ( r =0.929,P < 0.01 ).Conclusions IGF- Ⅰ R might be taken as a marker of clone cells in MDS for its trait to malignant proliferation.
