目的 提取胸腺素并检测药物指标,观察其临床应用和动物实验效果,探讨胸腺素对哮喘豚鼠肺组织Gα表达的影响。方法 采用酸碱沉淀加离心法制备胸腺素,经多肽定量、活性鉴定、细菌培养、毒性试验和热原实验,临床疗效观察,并以卵蛋白雾化吸入致敏法复制哮喘模型,Western blot分析法测定哮喘豚鼠肺组织Gα水平。结果 所制胸腺素符合中国药典标准。临床疗效确切,经治疗后,哮喘豚鼠肺组织Giα、Gqα水平显著降低(P<0.05),Gsα无显著变化。结论 采用酸碱沉淀加离心法可制备胸腺素,其治疗哮喘动物的机理可能是通过改变肺组织Gα水平来达到。

목적 제취흉선소병검측약물지표,관찰기림상응용화동물실험효과,탐토흉선소대효천돈서폐조직Gα표체적영향。방법 채용산감침정가리심법제비흉선소,경다태정량、활성감정、세균배양、독성시험화열원실험,림상료효관찰,병이란단백무화흡입치민법복제효천모형,Western blot분석법측정효천돈서폐조직Gα수평。결과 소제흉선소부합중국약전표준。림상료효학절,경치료후,효천돈서폐조직Giα、Gqα수평현저강저(P<0.05),Gsα무현저변화。결론 채용산감침정가리심법가제비흉선소,기치료효천동물적궤리가능시통과개변폐조직Gα수평래체도。
Objective To extract thymosin, detect the pharmaceutical indexes of it,observe the effect ofit in clinical application and animal test, and explore the influence of it on the expression of Ga protein in thelung tissue of guinea pig with asthma. Methods Thymosin was prepared by acid-base precipitation and cen-trifugation, and the pharmaceutical indexes of it were detected by quantification of polypeptide, identification ofactivity, bacterial culture, toxicity test and pyrogen test. The clinical effect of it was observed. Guinea-pig modelwith asthma was established by aerosol inhalation of egg albumin. The Ga protein level in the lung tissue ofguinea pig with asthma was analyzed by Western blot. Results The thymosin met the requirement of ChinesePharmacopeia. The clinical effect of it was definite. The Gia and Gqa levels in the lung tissues of guinea-pigswere significantly decreased after being treated with it (p<0.5). However, no significant change of Gsa levelwas observed. Conclusiou The thymosin prepared by acid-base precipitation and centrifugation can be appliedin clinic. The curative mechanism of it might be changing the Ga protein level in lung tissue.
