Nuclear imaging
目的 探讨IL-2作为自身免疫胰岛炎核素显像药物的可行性.方法 将38只雌性未发病NOD小鼠和26只Balb/c小鼠随机分为5个时间组,尾静脉注射酶法标记的125I-IL-2,相应时间处死后测定血液和腹腔各脏器放射性.另3只NOD小鼠在注射131I-IL-2 30 min后行SPEC-CT仪扫描显像.结果 注射后10至120 min,NOD小鼠胰腺135I-IL2聚集性明显高于Balb/c小鼠,且胰腺放射性与胰岛炎程度呈线性正相关(R2=0.614~0.776,P<0.05);但胰腺/周围组织放射性比值普遍较低;去除脾脏、肝脏和双肾,NOD小鼠胰腺方可显像.结论 125I-IL-2在自身免疫胰岛炎的NOD小鼠胰腺有相对高聚集性,但有较高放射性背景,影响局部显像效果.

Objective To evaluate the feasibility of ~ 125 Iodine -labeled IL-2 scintigraphic methods for the imaging of lymphocyte infiltration of the Langerhans′ islets. Methods ~ 125 I-IL2 was injected intravenously to pre-diabetic female NOD mice (aged 16-17 weeks)and female Balb/c mice as control. Animals were sacrificed at different time points and the radioactivity in different organs was measured.~ 131 I-IL-2 was injected iv to 3 pre-diabetic female NOD mice, and pancreas was imaged through SPECT after 30 minutes injection. Results The mean radio activities in the pancreas of NOD mice were significantly higher as compared to those of Balb/c mice at time points from 30 to 120 minutes (P<0.001), and it was positively correlated with insulitis score (R~2=0.614~0.776, P<0.03). The uptake ratios of pancreas over surrounding organs were almost all lower than 2.0. Pancreas was imaged only 30 minutes after ~ 131 I-IL-2 injection while spleen, liver and kidney were ablated. Conclusions The preliminary data show that the ~ 125 Iodine labeled IL-2 accumulates significantly in the pancreas of NOD mice, but it is needed to eliminate the higher radioactivity background in order to visualize the pancreas imaging.
