Sevoflurane%Drosophila melanogaster%K~+ currents
目的 以钾通道为切入点,利用膜片钳技术探讨吸入麻醉药的作用机制.方法 分离制备对吸入麻醉药敏感(S)、耐药(R)和野生(H)果蝇品系脑神经元,以膜片钳技术观察临床有效浓度七氟醚对其全细胞钾电流的影响.结果 1 MAC七氟醚使三品系Ⅲ型钾电流的Ipeak值显著增加:S(19±2)%>H(13±2)%>R(8±2)%;使钾电流的恢复曲线显著上移;不改变钾离子的平衡电位.2 MAC七氟醚使三品系Ⅲ型钾电流的Ipeak值显著增加:S(36±4)%>H(24±2)%>R(14±1)%.3 MAC的七氟醚能使Ⅲ型钾电流稳态失活曲线显著右移.结论 钾离子通道可能是吸入麻醉药的中枢作用位点之一.

Objective Drosophila was applied as animal model to investigate K~+ channels targets for volatile anesthetics with voltage-clamp recording technique. Methods Three strains of Drosophila with high (S), normal (H) and low (R) sensitivity to sevoflurane were developed by 33 generations" screening selection. Cerebral neurons from the three strains of Drosophila were obtained by acutely enzymatic digestion and incubation in fetal bovine serum. Whole-cell potassium currents were recorded using voltage-clamp recording technique. Sevoflurane was perfused to type Ⅲ neurons endowed with type Ⅲ whole-cell potassium currents consecutively with a Y-tube system. Results When treated with 1 or 2 MAC sevoflurane, Ipeak values of type Ⅲ currents were significantly potentiated in an order of S [(0. 19±0. 02) or (0. 36±0. 04)]>H[(0. 13±0. 02) or (0. 24±0. 02)]>R[(0. 08±0. 02) or (0. 14±0. 01)]. Effect of 1 MAC sevoflurane on the reversal potentials of type Ⅲ currents was not significantly different from the control The recovery curve of type Ⅲ currents was significantly upward shifted by 1 MAC sevoflurane. The steady-inactivation curve of type Ⅲ currents was significantly right-shifted by 3 MAC sevoflurane. Conclusion The results suggest that volatile anesthetics may act on K~+ channels in the mechanisms of anesthesia.
