目的 研究神经生长因子(NGF)对氧合血红蛋白(OxyHb)诱导的小鼠神经细胞凋亡的作用,进一步探讨OxyHb诱导细胞凋亡及NGF对神经元保护作用的可能机制.方法 蛛网膜下腔注射OxyHb建立蛛网膜下腔出血的动物模型,尾静脉注射NGF,TUNEL法检测神经细胞凋亡,免疫组织化学法检测Bcl-2、Bax、P75NTR和TrkA表达的情况.结果 注射OxyHb后出现神经细胞凋亡,Bcl-2表达降低,Bax和P75NTR表达增加,在NGF给药组,神经细胞凋亡数明显减少,Bcl-2表达增加.而Bax和P75NTR表达则明显降低.结论 小鼠局部脑组织蛛网膜下腔注射OxyHb可引起小鼠神经细胞发生凋亡,Bax和P75NTR表达增加可能是诱导凋亡的一个主要原因,而静脉注射NGF可以抑制OxyHb诱导的细胞凋亡,其机制可能是通过与受体结合,增加Bel-2蛋白表达,降低Bax和P75NTR表达水平以实现其保护作用.
目的 研究神經生長因子(NGF)對氧閤血紅蛋白(OxyHb)誘導的小鼠神經細胞凋亡的作用,進一步探討OxyHb誘導細胞凋亡及NGF對神經元保護作用的可能機製.方法 蛛網膜下腔註射OxyHb建立蛛網膜下腔齣血的動物模型,尾靜脈註射NGF,TUNEL法檢測神經細胞凋亡,免疫組織化學法檢測Bcl-2、Bax、P75NTR和TrkA錶達的情況.結果 註射OxyHb後齣現神經細胞凋亡,Bcl-2錶達降低,Bax和P75NTR錶達增加,在NGF給藥組,神經細胞凋亡數明顯減少,Bcl-2錶達增加.而Bax和P75NTR錶達則明顯降低.結論 小鼠跼部腦組織蛛網膜下腔註射OxyHb可引起小鼠神經細胞髮生凋亡,Bax和P75NTR錶達增加可能是誘導凋亡的一箇主要原因,而靜脈註射NGF可以抑製OxyHb誘導的細胞凋亡,其機製可能是通過與受體結閤,增加Bel-2蛋白錶達,降低Bax和P75NTR錶達水平以實現其保護作用.
목적 연구신경생장인자(NGF)대양합혈홍단백(OxyHb)유도적소서신경세포조망적작용,진일보탐토OxyHb유도세포조망급NGF대신경원보호작용적가능궤제.방법 주망막하강주사OxyHb건립주망막하강출혈적동물모형,미정맥주사NGF,TUNEL법검측신경세포조망,면역조직화학법검측Bcl-2、Bax、P75NTR화TrkA표체적정황.결과 주사OxyHb후출현신경세포조망,Bcl-2표체강저,Bax화P75NTR표체증가,재NGF급약조,신경세포조망수명현감소,Bcl-2표체증가.이Bax화P75NTR표체칙명현강저.결론 소서국부뇌조직주망막하강주사OxyHb가인기소서신경세포발생조망,Bax화P75NTR표체증가가능시유도조망적일개주요원인,이정맥주사NGF가이억제OxyHb유도적세포조망,기궤제가능시통과여수체결합,증가Bel-2단백표체,강저Bax화P75NTR표체수평이실현기보호작용.
Objectives To investigate the effect of nerve growth factor (NGF) on neuronal apoptosis and the expression of Bcl-2,Bax,P75NTR and TrkA which were induced by oxyhemoglobin, and to study the possible mechanism of apoptosis and the protective effect of NGF. Methods The oxyhemoglobin(OxyHb) was injected into the subarachnoid space of ICR mice, and NGF was injected into the body of mice in NGF group by intravenous after injury. TUNEL method was used to detect apoptosis and immunohistochemistry staining was used to detect the expression of Bcl-2, Bax , P75NTR and TrkA. Results After the injection of OxyHb, the TUNEL-positive neurons were seen in cortex, the expression of Bcl-2 was decreased, while the expression of P75NTR and Bax were increased. In NGF group, the amount of TUNEL positive cells was obviously decreased, and the expression of Bcl-2 was significantly increased, while the expression of Bax and P75NTR positive cells were obviously decreased. The expression of TrkA has no significantly difference in both groups. Conclusion OxyHb in subarachnoid space can induced apoptosis in neuronal cells of mouse in vivo. The increased expression of P75NTR and Bax after OxyHb injection into subarachnoid space may be one of the main reasons for apoptosis after subarachnoid hemorrhage. Intravenous injection of NGF might has protective effect on neuronal apoptosis after subaraehnoid hemorrhage via regulation the expression of Bcl-2, Bax and P75NTR through combination with NGF receptor.
