制备了人胶质瘤细胞SWO-38条件培养基(SWO-38 CM),经超滤浓缩,截留分子量大于10 kD的SWO-38 CM,并运用快速自动比色微量分析法检测其对体外培养的SD大鼠小脑皮质神经元的作用.结果表明,分子量大于10 kD的SWO-38 CM对神经元的作用,与对照组比较有明显的神经营养活性.分子量大于10 kD的SWO-38 CM经Sephacry1S-200-HR凝胶层析并结合生物活性鉴定,获得了具有神经营养活性的第Ⅱ峰洗脱液.
製備瞭人膠質瘤細胞SWO-38條件培養基(SWO-38 CM),經超濾濃縮,截留分子量大于10 kD的SWO-38 CM,併運用快速自動比色微量分析法檢測其對體外培養的SD大鼠小腦皮質神經元的作用.結果錶明,分子量大于10 kD的SWO-38 CM對神經元的作用,與對照組比較有明顯的神經營養活性.分子量大于10 kD的SWO-38 CM經Sephacry1S-200-HR凝膠層析併結閤生物活性鑒定,穫得瞭具有神經營養活性的第Ⅱ峰洗脫液.

Human glioma cell line (SWO-38) conditioned medium (SWO-38 CM)was prepared,ultratiltrated and concentrated, so the fraction of molecular weight (MW)>10 kD from SWO-38CM was obtained. The effect of SWO-38 CM fractions (MW>10 kD and MW<10kD)on theactivity of cultured cerebellar cortical neurons of Spraque-Dawley rat was measured by a rapidautomated colorimetric microassay, respectively. The data indicated that SWO-38 CM fraction(MW>10 kD) possessed significant neurotrophic activity to the neurons, as compared to thecontrol (P<0.05). Additionally, the active fraction of SWO-38 CM was purified further bySephacryl S-200-HR gel chromatography and peak Ⅱ was only identified to possess neurotrophicactivity.
