Diabetic Nephropathies%Proteinuria%Angiotensin - Converting Enzyme Inhibitors
目的 研究厄贝沙坦治疗糖尿病肾病(DN)蛋白尿作用机制.方法 应用链脲佐菌素建立大鼠糖尿病肾病模型,成模大鼠随机分为模型对照组(M)、厄贝沙坦干预组(I),另设正常对照组(C).各组干预12周后,分别检测:①血糖、血肌酐、24h尿蛋白定量等;②肾脏病理、电镜以及免疫组化分析;③实时定量RT-PCR分析.结果 与正常组比较,模型组大鼠24h尿蛋白含量显著增加,肾小球基底膜增厚,足细胞nephrin、podocin表达显著减少,通过厄贝沙坦干预治疗,上述指标改善(P<0.05),且蛋白尿的程度与nephrin、podocin表达呈负相关(P<0.05).结论 厄贝沙坦可通过拮抗DN大鼠的足细胞损害;减少其尿蛋白.
目的 研究阨貝沙坦治療糖尿病腎病(DN)蛋白尿作用機製.方法 應用鏈脲佐菌素建立大鼠糖尿病腎病模型,成模大鼠隨機分為模型對照組(M)、阨貝沙坦榦預組(I),另設正常對照組(C).各組榦預12週後,分彆檢測:①血糖、血肌酐、24h尿蛋白定量等;②腎髒病理、電鏡以及免疫組化分析;③實時定量RT-PCR分析.結果 與正常組比較,模型組大鼠24h尿蛋白含量顯著增加,腎小毬基底膜增厚,足細胞nephrin、podocin錶達顯著減少,通過阨貝沙坦榦預治療,上述指標改善(P<0.05),且蛋白尿的程度與nephrin、podocin錶達呈負相關(P<0.05).結論 阨貝沙坦可通過拮抗DN大鼠的足細胞損害;減少其尿蛋白.
목적 연구액패사탄치료당뇨병신병(DN)단백뇨작용궤제.방법 응용련뇨좌균소건립대서당뇨병신병모형,성모대서수궤분위모형대조조(M)、액패사탄간예조(I),령설정상대조조(C).각조간예12주후,분별검측:①혈당、혈기항、24h뇨단백정량등;②신장병리、전경이급면역조화분석;③실시정량RT-PCR분석.결과 여정상조비교,모형조대서24h뇨단백함량현저증가,신소구기저막증후,족세포nephrin、podocin표체현저감소,통과액패사탄간예치료,상술지표개선(P<0.05),차단백뇨적정도여nephrin、podocin표체정부상관(P<0.05).결론 액패사탄가통과길항DN대서적족세포손해;감소기뇨단백.
Objectives This research aims to study the mechanism of action of Irbesartan in treating proteinuria in diabetic nephropathy (DN).Methods DN rat models were prepared with an intraperitoneal injection,which were then randomly divided into model group ( M group) and Irbesartan group ( I group).There was also a normal control group (C group).After 12 -week intervention,the following tests and measures were carried out for each group:(1) Blood glucose,serum creatinine (Cr),blood urea nitrogen (BUN) and 24 -hour urine protein level tests ; ( 2 ) Pathological tests,immunohistochemistry ( IHC ) and electron microscopy analyses; ( 3 ) Quantitative real - time RT - PCR.Results Noticeably increased 24 - hour urine protein levels and glomerular basement membrane (GBM) thickness and noticeably decreased expression of nephrin and podocin in podocytes were found in M group compared to C group.Irbesartan intervention improved the above indicators ( P < 0.05 ).Furthermore,the severity of proteinuria was found to be negatively correlated with nephrin expression and podocin expression( P <0.05).Conclusions Irbesartan is able to lower the urine protein level by inhibiting damage to podocytes in DN rats.
