目的 观察异丙酚预处理后Gq/11蛋白在急性肺损伤(acute lung injury,ALI)大鼠脑组织中的动态变化,探讨异丙酚是否通过影响Gq/11蛋白的表达对脑组织起保护作用.方法 用尾静脉注射油酸法复制ALI大鼠模型,将24只健康雄性Wistar大鼠随机分为对照组、ALI组和异丙酚预处理组.检测各组大鼠血气(pH值、PaO2、PaCO2)、平均动脉血压(MABP)以及血浆和脑组织乳酸脱氢酶(LDH)、肌酸激酶(CK)、丙二醛(MDA).Western blot检测各组大鼠脑组织中的Gq/11蛋白含量.结果 异丙酚预处理组Gq/11蛋白含量为(133.89±21.59)%,较对照组增加(33.89±21.59)%(P<0.01),但低于ALI组[(165.84±30.27)%,P<0.05].异丙酚预处理组及ALI组PaO2较对照组明显降低(P值均<0.01),但异丙酚预处理组高于ALI组(P<0.01).异丙酚预处理组及ALI组MABP均较对照组降低(P值均<0.01),异丙酚预处理组低于ALI组(P<0.01).异丙酚预处理组脑组织LDH和CK活性分别为(1.51±0.58)U/mg、(35.83±7.09)U/mg,MDA含量为(4.70±1.59)U/mg,均低于ALI组[分别为(2.38±0.72)U/mg(P<0.01),(44.31±8.15)U/mg(P<0.01),(6.78±1.38)U/mg(P<0.05)].血浆LDH和CK活性、MDA含量呈现与之相似的变化趋势.结论 异丙酚预处理能减轻ALI时脑的损伤程度,其保护作用可能与下调Gq/11蛋白有关.

Objective To observe the alteration of Gq/11 protein expression in the brain of rats with acute lung injury(ALI)after propofol pre-treatment and to explore whether it plays a protective role in brain injury during ALI.Methods ALI rat model was reproduced by intravenous injection of oleic acid.Twentyfour male Wister rats were randomly divided into three groups:control group,ALI group and propofol group.Blood gas indexes(pH,PaO2,PaCO2),mean arterial blood pressure(MABP)and MDA,LDH,CK of plasma and brain were measured.Gq/11 protein concentration was examined by Western blot.Results Gq/11protein concentration in the brain of propofol group[(133.89±21.59)%]was increased compared with control group(P<0.01),and it was decreased compared with ALI group[(165.84±30.27)%,P<0.053.PaO2 of propofol groUp and ALI group were lower than that of control group(all P<0.01),and PaO2 of propofol group was higher than that of ALI group(P<0.01).MABP of propofol group and ALI group obviously reduced(all P<0.01),and MABP of propofol group was lower than that of ALI group(P<0.01).LDH activity[(1.51±0.58)U/mg],CK activity[(35.83±7.09)U/mg]and MDA content[(4.70±1.59)U/mg]in homogenato of propofol group were lower than those in ALI group[(2.38±0.72)U/mg(P<0.01),(44.31±8.15)U/mg(P<0.01),(6.78±1.38)U/mg(P<0.05)].The levels of LDH,CK and MDA in plasma were same as in brain.Conclusions Pre-treatment with propofol can relieve the harm,which has some relation with reducing Gq/11 protein concentration.
