目的:探讨罗格列酮对脂肪细胞增殖和胰岛素抵抗的影响.方法:培养3T3-L1前脂肪细胞。用四甲基偶氮唑盐(MTT)方法检测3T3-L1前脂肪细胞及脂肪细胞的增殖情况,同时采用地塞米松诱导3T3-L1脂肪细胞建立胰岛素抵抗模型,检测罗格列酮对细胞培养基中葡萄糖浓度的影响.结果:在DMEM高糖培养基中。罗格列酮可促进3T3-L1脂肪细胞增殖,抑制3T3-L1前脂肪细胞增殖;使3T3-L1前脂肪细胞及3T3-L1脂肪细胞葡萄糖消耗量增加;罗格列酮能明显增加胰岛素抵抗3T3-L1脂肪细胞培养基中的葡萄糖消耗量,增强对胰岛素的敏感性.结论:罗格列酮可以促进脂肪细胞的增殖,抑制前脂肪细胞的增殖,罗格列酮对地塞米松诱导3T3-L1脂肪细胞的胰岛素抵抗具有明显的改善作用.

목적:탐토라격렬동대지방세포증식화이도소저항적영향.방법:배양3T3-L1전지방세포。용사갑기우담서염(MTT)방법검측3T3-L1전지방세포급지방세포적증식정황,동시채용지새미송유도3T3-L1지방세포건립이도소저항모형,검측라격렬동대세포배양기중포도당농도적영향.결과:재DMEM고당배양기중。라격렬동가촉진3T3-L1지방세포증식,억제3T3-L1전지방세포증식;사3T3-L1전지방세포급3T3-L1지방세포포도당소모량증가;라격렬동능명현증가이도소저항3T3-L1지방세포배양기중적포도당소모량,증강대이도소적민감성.결론:라격렬동가이촉진지방세포적증식,억제전지방세포적증식,라격렬동대지새미송유도3T3-L1지방세포적이도소저항구유명현적개선작용.
Objective:To investigate the influence of rosiglitazone on proliferation of 3T3 -L1 adipocytes and insulin resistance. Methods 3T3 - L1 preadipocytes were cultured, the proliferation of 3T3 - L1 preadipocytes was detected by MTT method. Insulin resistant 3T3 - L1 adipocytes cell model was induced by dexamethasone and the change of glucose concentration in cell culture was determined after rosiglitazone treatment. Results In the high glucose DMEM culture media, MTT method showed that the absorbance at 570nm of 3T3 -L1 preadipocytes were increased and that of 3T3 - L1 adipocytes were decreased. rosiglitazone significantly increased glucose consumption in 3T3 - L1 preadipocytes and adipocytes culture with a concentration - dependent effect. Rosiglitazone improved the sensitivity of 3T3 - L1 adipocytes to insulin. Conlcusions Rosiglitazone can promote the proliferation of 3T3 - L1 adipocytes and inhibited the proliferation of 3T3 - L1 preadipocytes, rosiglitazone can significantly improve insulin resistance induced by dexamethaone.
