目的:探讨大鼠 L5神经根受压后,益气化瘀方在神经再生修复过程中对神经肌肉接头部神经元的作用和表现. 方法:采用多克隆蛋白基因产物 9.5( protein gene product 9.5,PGP 9.5)作为神经元标记,大鼠 L5神经根受压后,给予益气化瘀方灌胃,分别于术后 10,20,30和 60 d取比目鱼肌,用免疫组织化学方法结合激光共聚焦扫描显微技术,观察神经肌肉接头部末梢神经再生修复过程.α-bungarotoxin(α-BTX)荧光结合剂显示运动终板, NIH图像分析技术测定末梢神经与运动终板重叠面积. 结果:肌肉失神经支配后,益气化瘀方组的末梢神经在神经肌肉接头部的聚集,出芽及延伸均明显早于对照组.神经再生修复期间,末梢神经与运动终板的重叠速度和范围以及神经肌肉接头的再构筑,益气化瘀方组亦同样明显优于对照组. 结论:益气化瘀方能促进神经元的增生及提高其再生功能,加快神经肌肉接头的重建及明显缩短神经再生修复的进程.

AIM:To study the effect of Yiqi Huayu Fang on neurons at neuromuscular junctions during regeneration and repair of nerve after L5 nerve root compressed in rats. METHODS:Using polyclonal protein gene product 9.5(PGP 9.5) as a neuron mark,the soleus muscles of the rats were obtained after the Yiqi Huayu Fang was given through stomach perfusion at 10,20,30 and 60 d after L5 nerve root compressed.The process of peripheral nerve regeneration and repair at neuromuscular junctions was observed with immunohistochemistry and confocal laser scanning techniques.The motion endplates were visualized with fluorescein-conjugated α-bungarotoxin(α-BTX).The overlapping areas of peripheral nerve and motion endplates were measured with NIH image technique. RESULTS:After denervation of muscle,the aggregation,budding and extension of peripheral nerve at neuromuscular junctions in Yiqi Huayu Fang group(trial group) were obviously earlier than those in control group.The overlapping speed and range of peripheral nerve with motion endplates and reformation of neuromuscular junctions during regeneration and repair of nerve in trial group were significantly better than those in control group. CONCLUSION:Yiqi Huayu Fang can promote proliferation of neurons and enhance their regeneration function.Meanwhile,it can accelerate the reformation of neuromuscular junction and remarkably shorten the process of regeneration and repair of nerve.
