目的 观察中药益坤宁对围绝经期大鼠海马芳香化酶表达的影响,探讨益坤宁治疗围绝经期综合征的作用机制.方法 采用自然老化法建立围绝经期大鼠模型.选择处于围绝经期的雌性Wistar大鼠(12-14月龄)30只,随机分为围绝经期对照组、两药对照组和中药益坤宁组,分别以生理盐水、利维爱和益坤宁连续灌胃4周;另取5月龄大鼠10只作为青年对照组,生理盐水连续灌胃4周.通过HE染色法观察大鼠海马神经元形态学变化,免疫组化法检测大鼠海马芳香化酶蛋白的表达,RT-PCR检测各组大鼠海马芳香化酶mRNA的表达.结果 HE染色显示中药益坤宁组大鼠海马神经元胞体和胞核较围绝经期对照组显著增大,神经元密度增加(P<0.01).免疫组化结果 显示,芳香化酶蛋白表达于各组大鼠海马的神经元内,中药益坤宁组大鼠芳香化酶蛋白的表达明显高于围绝经期对照组(P<0.01);RT-PCR结果 显示,青年对埘照组、中药益坤宁组和西药对照组芳香化酶mRNA的表达均高于围绝经期对照组(P<0.01),且mRNA表达的结果 与蛋白表达的结果 一致.结论 益坤宁能够促进海马神经元的生长,上调围绝经期大鼠海马芳香化酶蛋白和mRNA的表达,可能是其治疗围绝经期综合征的机制之一.

목적 관찰중약익곤저대위절경기대서해마방향화매표체적영향,탐토익곤저치료위절경기종합정적작용궤제.방법 채용자연노화법건립위절경기대서모형.선택처우위절경기적자성Wistar대서(12-14월령)30지,수궤분위위절경기대조조、량약대조조화중약익곤저조,분별이생리염수、리유애화익곤저련속관위4주;령취5월령대서10지작위청년대조조,생리염수련속관위4주.통과HE염색법관찰대서해마신경원형태학변화,면역조화법검측대서해마방향화매단백적표체,RT-PCR검측각조대서해마방향화매mRNA적표체.결과 HE염색현시중약익곤저조대서해마신경원포체화포핵교위절경기대조조현저증대,신경원밀도증가(P<0.01).면역조화결과 현시,방향화매단백표체우각조대서해마적신경원내,중약익곤저조대서방향화매단백적표체명현고우위절경기대조조(P<0.01);RT-PCR결과 현시,청년대시조조、중약익곤저조화서약대조조방향화매mRNA적표체균고우위절경기대조조(P<0.01),차mRNA표체적결과 여단백표체적결과 일치.결론 익곤저능구촉진해마신경원적생장,상조위절경기대서해마방향화매단백화mRNA적표체,가능시기치료위절경기종합정적궤제지일.
Objective To investigate mechnism of traditional Chinese medicine Yikunning treating perimenopausal syndrome,the effects of Yikunning on the expression of aromatase in rat hippocampal during perimenopausal period.Methods Thirty female Wistar rats during perimenopausal period were selected by unforced aging.Then the rats were divided into 3 groups randomly,perimenopausal period model group,western medicine control group and Chinese medicine Yikunning group.Ten young female Wistar rats were selected as young control group.Intragastric administrations were conducted for 4 weeks once daily continuously.The morphological changes of the neurons in rat hippocampus were detected by HE staining.The expression of aromatase in hippocampus of rats was detected by immunohistochemistry.The expression of aromatase mRNA in hippocampus of rats was detected by RT-PCR.Results After HE staining,in hippocampus of Chinese medicine Yikunning group,the body and nucleus of the neurons became larger than those in perimenopausal period model controls,and the neurons density also strengthened.which approaches to young controls and western medicine Livial controls(P<0.01).Compared with young controls,the expression of aromatase mRNA in hippocampus of perimenopausal period model controls decreased significantly,while the expression of aromatase in hippocampus of Chinese medicine Yikunning group increased significantly.The expressions of aromatase mRNA in the young controis,Chinese medicine Yikunning group and western medicine Livial controls were higher than that in perimenopausal period model controls(P<0.01),which were consisted with the results aromatase protein.Conclusion Yikunning could improve the growth and development of neurons in hippocampus,and up-regulate the expression of aromatase mRNA and protein in rat hippocampus during perimenopausal period,which may be one of the mechanism of Yikunning to treat perimenopausal syndrome.
