目的:为进一步提高产乳糖酶酵母Kluyveromyces fragilis(K.fragilis)株的酶产量,对该菌株的培养产酶发酵条件进行优化试验。方法:采用增殖和诱导二步培养法培养酵母K.fragilis株,并采用单因素试验方法分析碳源、氮源对酵母诱导产酶的影响。结果:确定了K.fragilis酵母株诱导培养的较佳配方,当乳糖浓度为120g/L,醋酸铵浓度为5g/L,pH为6.7时,单位酶产量达122.4IU/ml培养液。结论:该菌株生长产酶情况良好,具有较高的应用价值。

목적:위진일보제고산유당매효모Kluyveromyces fragilis(K.fragilis)주적매산량,대해균주적배양산매발효조건진행우화시험。방법:채용증식화유도이보배양법배양효모K.fragilis주,병채용단인소시험방법분석탄원、담원대효모유도산매적영향。결과:학정료K.fragilis효모주유도배양적교가배방,당유당농도위120g/L,작산안농도위5g/L,pH위6.7시,단위매산량체122.4IU/ml배양액。결론:해균주생장산매정황량호,구유교고적응용개치。
Objective: To study the optimal condition on lactase production by yeast Kluyveromyces fragilis(K.fragilis) to increase the enzyme productivity.Methods: With separated proliferation and induction culture,the effe cts of carbon source and nitrogen source on yeast proliferation and lactase indu ction were studied by the method of monofactorial analysis.Results: The optimum medium for the productivity of lactase by K.f ragilis was determined.Maximal enzyme activity (122.4 IU/ml) was reached when the concentration of lactose was 12%,ammonium acetate was 0.5% and pH 6.7.Conclusion: The results of this experiment demonstrate that this str ain will be good for industrial production.
