目的探讨P14、P16在宫颈癌组织中的表达及其与临床病理参数的相关性.方法采用免疫组织化学链霉素抗生素-过氧化酶连接法(S-P法)检测P14、P16在56例宫颈鳞癌组织、20例CIN Ⅲ组织和30例正常宫颈组织中的表达情况.结果 P14和P16蛋白在正常宫颈组织中的阳性表达率明显低于CIN Ⅲ组织和宫颈癌组织,差异有显著性(P＜0.05).P14蛋白的表达与浸润深度、病理分级及淋巴转移有关(P＜0.05),与临床分期、年龄、肿瘤大小或脉管有无癌栓无关(P＞0.05)；P16蛋白的表达与病理分级及淋巴转移有关(P＜ 0.05),与临床分期、年龄、肿瘤大小、浸润深度或脉管有无癌栓无关(P＞0.05).结论 P14、P16基因的高表达发生在宫颈癌和癌前病变阶段,对宫颈癌的诊断及预后判断有一定价值,故可作为肿瘤标记物用于宫颈癌的临床筛查及诊断.

Objective To explore the expressions of P14 and P16 in cervical carcinoma tissue and its relationship with clinical pathological-parameters.Methods Expressions of P14 protein and P16 in 56 cases' squamous cervical arcinoma tissue,20 cases' CIN Ⅲ tissue and 30 cases' normal cervical tissue were detected with S-P immunochemistry technology.Results The positive expression rates of P14 and P16 proteins were significantly lower in normal cervical tissue than in the CIN Ⅲ and squamous cervical carcinoma( P＜ 0.05 ).The expression of P14 in squamous cervical carcinoma related to tumor infiltrate deepness,pathological stage and lymph node metastasis ( P ＜ 0.05 ) but not related to clinical stage,age,tumor size and cancer embolus( P＞ 0.05 ).The expression of P16 in squamous cervical carcinoma related to pathological stage and lymph node metastasis ( P ＜ 0.05 ) but not related to clinical stage,age,tumor size, tumor infiltrate deepness and cancer embolus ( P ＞ 0.05 ).Conclusions P14 and P16 expess highly in CIN and cervical carcinoma,which has value in the diagnosis and prognostic valuation of cervical carcinoma.Consequently,both of them can be used as biomarkers for the screening and diagnosis of cervical carcinoma.
