目的 探讨Livin、p53在不同级别人脑胶质瘤中的表达及其与肿瘤恶性程度之间的关系.分析二者在胶质瘤细胞凋亡过程中的信号转导机制.方法 应用免疫组织化学技术检测Livin、p53蛋白在41例人脑胶质瘤和10例正常脑组织中的表达.结果 Livin、p53蛋白在10例正常脑组织中均不表达,在41例胶质瘤组织中表达率分别为87.8%和41.5%.Livin和p53蛋白在低级别胶质瘤中表达较低,在高级别胶质瘤中表达较高,两组比较差异有统计学意义(P<0.05).Spearman等级相关分析显示,Livin蛋白与p53蛋白表达呈正相关(r=1.000,P<0.01).结论 Livin和p53在人脑胶质瘤组织中表达上调,细胞凋亡受到抑制,引起肿瘤恶性增殖,与胶质瘤病程发展、恶性程度明显相关,这可能是胶质瘤恶性增殖的一个机制.

Objective To investigate the expressions of Livin and p53 proteins in human glioma of different histological grades,and to analyze their relationships with degree of malignancy,and the signal transduction mechanism of inhibiting cell apoptosis. Methods Livin and p53 protein expression levels were measured by immunohistochemistry technique in 41 human gliomas and 10 normal brain tissues respectiveIy. Results The positive expression rates of Livin and p53 were 87.8% and 41.5%in gliomas, respectively, while there was no expression in 10 normal brain tissues.The expressions of Livin and p53 in high-grade gliomas were significantly higher than those in low-grade glioma,and the expressions were significantly related to the pathological grade(p<0.05).According to Spearman analysis, the correlation coefficient r=1.000, P<0.01, this showed Livin expression had a positive correlation with p53 expression. Conclusions The expressions of Livin and p53 are up-regulated in human glioma,which inhibits cell apoptosis and leads to malignant proliferation of tumor.Livin and p53 might play important roles in the oncogenesis and progression of human glioma. Livin expression is in accordance with p53 mutation in the tumorigenesis of gliomas. This may be one of the mechanisms of maligllant proliferation of tumor.
