目的探讨骨质疏松症患者人成骨细胞理想的培养和鉴定方法.方法在关节手术时取骨质疏松症患者股骨头松质骨,以改良酶消化法作人成骨细胞体外分离培养,成骨细胞的鉴定采用改良Gomori碱性磷酸酶(ALP)钙钴法染色.结果改良酶消化法获得的成骨细胞数量多,纯度高,密度均匀一致.结论采用改良酶消化法获取原代细胞,经培养、传代后,可得到较为稳定、纯化的骨质疏松患者人成骨细胞,此细胞适于做体外实验研究模型.

Objective To explore the ideal method of human osteoporosis osteoblast culture in vitro and identification.Methods Cancellous bone was obtained from femoral head of patients with osteoporosis undergoing joint arthroplasty.Human osteoblasts isolated from improved collagenase-treated bone explants were cultured in vitro.Identification of osteoblasts is based on morphology and improved Gomori alkaline phosphatase (ALP) Ca-Co staining.Results More osteoblasts were acquired by improved enzyme digestion,with features of high purity and average density,and most of them are in the same cell cycle.Conclusion Human primary osteoblasts by improved enzyme digestion,after being cultured,can develop into stable and purified human osteoporosis osteoblasts and act as in vitro model.
