以壳聚糖为研究对象,对其进行精氨酸改性研究。以1-(3-二甲氨基丙基)-3-乙基碳二亚胺(EDC)为引发剂,N-羟基硫代琥珀酰亚胺(NHS)为催化剂,将精氨酸(Agrinine)接枝到壳聚糖上。通过控制精氨酸的加入量,反应温度等条件,从而得到不同精氨酸取代度(DS)的壳聚糖-精氨酸(CS-N-Arginine,CA)。CA的化学结构和热稳定性是通过红外分析、差示扫描量热分析表征。Arg-CS有望成为一种高效、安全的壳聚糖基非病毒载体。

In this experiment,chitosan used as target polymer and was modified with arginine.1-(3-dimethylamino propyl)-3-ethyl carbodiimide(EDC) as initiator and N-hydroxy succinimide thiosulfate(NHS) as the catalyst,the arginine was grafted to chitosan.A series of arginine modified chitosan(CS-N-Arginine,CA) with different substitution degree(DS),were prepared by controlling the amount of arginine,reaction temperature and other conditions.Chemical structure and thermal stability of CA were characterized through infrared analysis and differential scanning calorimetry characterization.Arg-CS was expected to be an efficient and safe chitosan-based non-viral vector.
