目的 观察长链非编码RNA(lncRNA)在正常肝组织及肝癌中表达谱的变化.方法 利用lncRNA芯片技术检测人正常肝脏、肝癌组织LncRNA表达谱的差异,经过对原始数据进行预处理、均一化后,筛选出差异表达LncRNA,进行分析.结果 与正常肝脏组织比较,其中2倍以上变化并且差异有统计学意义(P<0.05)的lncRNA,认为是差异表达的lncRNA,2倍以上变化的共837条,占所有lncRNA的4.30%;2倍以上升高的共325条;2倍以上降低的共512条;5倍以上升高为共47条;5倍以上降低的共69条;10倍以上升高为共7条;10倍以上降低的共6条.结论 肝癌与正常肝脏组织比较,LncRNA表达谱发生了显著变化,差异性lncRNAs中,可能包括相当一部分的促癌lncRNA和抑癌lncRNA,参与肝癌分子调节机制的关键环节.

Objective To analyze the expression profile variation of long non-coding RNA (IncRNA) in normal hepatic tissue and hepatocellular carcinoma.Methods Use lncRNA microarray technology to inspect the difference of lncRNA expression profile between normal human liver and hepatocellular carcinoma,screen out the lncRNA with differential expression after pretreatment and homogenization of raw data,and make hierarchical clustering and volcano scatter diagram analysis.Results Compare with normal hepatic tissue,and 837 lncRNA which have more than 2 times variation and significant difference (P <0.05 ) by statistical analysis are regarded as lncRNA with differential expression,accounting for 4.30% of all IncRNA ;while 325 increase more than 2 times and 512 reduce more than 2 times;47 increase more than 5 times;69 reduce more than 5 times;7 increase more than 10 times;and 6 reduce more than 10 times.Conclusion LncRNA expression profile of hepatoeellular carcinoma changes significantly in comparison with normal hepatic tissue.The different lncRNAs may include a fair proportion of cancer-promoting IncRNA and cancer-inhibiting lncRNA,participating in the critical section of regulatory mechanism in liver cancer molecule.
