目的:研究maspin、p53及skp2蛋白在结直肠肿瘤组织中的表达及临床病理意义.方法:采用免疫组织化学法检测maspin、p53及skp2在50例结直肠腺癌,20例结直肠腺瘤、20例正常结直肠粘膜组织中的表达.结果:maspin蛋白在结直肠腺瘤和正常结直肠粘膜组织中的表达均高于结直肠腺癌(p<0.05);无淋巴结转移组maspin表达率高于淋巴结转移组(p<0.05);Dukes C,D期结直肠癌组织maspin表达率低于Dukes A、B期(p<0.05).在结直肠腺癌及腺瘤中maspin与p53的表达均呈负相关(P<0.01),然而在skp2与maspin、skp2与p53之间没有发现相关性(P>O.05).结论 :maspin的表达可能受p53基因的调控.联合检测maspin、p53及skp2在结直肠肿瘤中表达水平,对判定结直肠癌恶性程度及预后可能具有重要参考价值.

Objective: To investigate the role of maspin,p53 and Skp2 expression in the progression of colorectal tumors,and to understand their clinicopathological significance. Methods: The expression of maspin,p53 and Skp2 in colorectal adenocarcinoma(n=50), adenoma(n=20)and normal tissue(n=20)were detected by immunohistochemistry and compared with the clinicopathological tumor parameters. The relationship among the expression of maspin,p53,and Skp2 was also analyzed in colorectal tumors.Results: Maspin expression was much lower in colorectal adenocarcinoma than in adenoma and normal tissues(P<0.05).Maspin expression showed a negative association with lymphatic metastasis and higher Dukes stage(P<0.05).Expression of maspin protein was negatively correlated with p53 expression(p<0.01).However,there was no significant correlation between expression of Skp2 and p53 or maspin(P>0.05).Conclusion: Maspin might be regulated by the p53 pathway.Combined detection of the expression of maspin,p53,and skp2 may be useful for evaluating its infiltration,metastasis and prognosis.
