目的 探讨不同剂量促红细胞生成素(EPO)减轻肾缺血再灌注损伤(IRI)后肾间质纤维化的作用及其机制.方法 通过单侧肾缺血再灌注肾损伤模型造成小鼠患侧肾间质纤维化,随机分为假手术组、模型组、高剂量和低剂量EPO给药组,术后28 d处死各组小鼠,取患侧肾行组织学检查,并用免疫组织化学法或Westen blot检测肾组织中胶原-Ⅳ( COL-Ⅳ)、结缔组织生长因子(CTGF)、骨桥蛋白(OPN)及单核细胞趋化因子-I(MCP-1)的水平.结果 模型组出现了明显的间质纤维化,两给药组肾间质纤维化与模型组比较有所减轻,COL-IV、CTGF、OPN及MCP-1的水平明显减低,高低剂量EPO组之间差异无统计学意义(P>0.05).结论 EPO可以通过影响细胞因子表达、抑制炎性细胞迁移来减轻细胞外胶原的积聚,从而减轻间质纤维化,发挥肾保护作用,高低剂量EPO具有相同的治疗效果.
目的 探討不同劑量促紅細胞生成素(EPO)減輕腎缺血再灌註損傷(IRI)後腎間質纖維化的作用及其機製.方法 通過單側腎缺血再灌註腎損傷模型造成小鼠患側腎間質纖維化,隨機分為假手術組、模型組、高劑量和低劑量EPO給藥組,術後28 d處死各組小鼠,取患側腎行組織學檢查,併用免疫組織化學法或Westen blot檢測腎組織中膠原-Ⅳ( COL-Ⅳ)、結締組織生長因子(CTGF)、骨橋蛋白(OPN)及單覈細胞趨化因子-I(MCP-1)的水平.結果 模型組齣現瞭明顯的間質纖維化,兩給藥組腎間質纖維化與模型組比較有所減輕,COL-IV、CTGF、OPN及MCP-1的水平明顯減低,高低劑量EPO組之間差異無統計學意義(P>0.05).結論 EPO可以通過影響細胞因子錶達、抑製炎性細胞遷移來減輕細胞外膠原的積聚,從而減輕間質纖維化,髮揮腎保護作用,高低劑量EPO具有相同的治療效果.
목적 탐토불동제량촉홍세포생성소(EPO)감경신결혈재관주손상(IRI)후신간질섬유화적작용급기궤제.방법 통과단측신결혈재관주신손상모형조성소서환측신간질섬유화,수궤분위가수술조、모형조、고제량화저제량EPO급약조,술후28 d처사각조소서,취환측신행조직학검사,병용면역조직화학법혹Westen blot검측신조직중효원-Ⅳ( COL-Ⅳ)、결체조직생장인자(CTGF)、골교단백(OPN)급단핵세포추화인자-I(MCP-1)적수평.결과 모형조출현료명현적간질섬유화,량급약조신간질섬유화여모형조비교유소감경,COL-IV、CTGF、OPN급MCP-1적수평명현감저,고저제량EPO조지간차이무통계학의의(P>0.05).결론 EPO가이통과영향세포인자표체、억제염성세포천이래감경세포외효원적적취,종이감경간질섬유화,발휘신보호작용,고저제량EPO구유상동적치료효과.
Objective To investigate the protective effect and possible mechanism of erythropoietin (EPO) on interstitial fibrosis of kidneys in mouse after ischemia reperfusion injury (IRI).Methods The tubulointerstitial fibrosis model via renal IRI was established.All mice were randomly divided into four groups:sham-operated,model group,high and low doses EPO treatment group.Mice were sacrificed at day 28 after operation.The renal morphological changes were observed by Periodic Acid-Schiff stain.The expression of COL-IV,CTGF,OPN and MCP-1 in the kidney were assessed by immunchistochemistry or Western blotting.Results The tubulointerstitial fibrosis and the levels of COL-IV,CTGF,OPN and MCP-1 in model group increased remarkably comparing in the sham-operated group.In comparison with the model group,the tubulointerstitial fibrosis and the levels of them were significantly reduced in EPO treatment groups.There was no difference between high and low dos EPO treatment group.Conclusion Erythropoietin may alleviate the renal interstitial fibrosis through influence of the expression of the cytokine,inhibition of the migration of inflammatory cells and followed decrease of the accumulation of the extracellular collagen.There was a same therapeutic effect in both high and low dos EPO treatment groups.
