Hepatitis B vaccine%Enzyme tests
目的 建立免疫复合型增效乙型肝炎疫苗细胞免疫检测方法并与常规乙型肝炎疫苗进行比较。方法增效乙型肝炎疫苗肌内免疫BALB/c小鼠,Elispot方法检测小鼠脾脏淋巴细胞分泌IFN-γ的细胞频数(SFC),优化实验条件:免疫剂量、检测时间和特异性刺激物及刺激浓度。同时将增效乙型肝炎疫苗与常规乙型肝炎疫苗进行比较。结果5μg免疫组IFN-γ SFC明显高于2μg免疫组,初次免疫后3周IFN-γ SFC开始下降,多肽刺激IFN-γ SFC高于蛋白刺激,多肽浓度为2μg即可充分刺激斑点形成;增效乙肝疫苗免疫组IFN-γ SFC高于常规乙型肝炎疫苗组。结论 初步建立了免疫复合型增效乙型肝炎疫苗细胞免疫检测方法并证明其重复性良好,其细胞免疫水平高于常规乙型肝炎疫苗。

목적 건립면역복합형증효을형간염역묘세포면역검측방법병여상규을형간염역묘진행비교。방법증효을형간염역묘기내면역BALB/c소서,Elispot방법검측소서비장림파세포분비IFN-γ적세포빈수(SFC),우화실험조건:면역제량、검측시간화특이성자격물급자격농도。동시장증효을형간염역묘여상규을형간염역묘진행비교。결과5μg면역조IFN-γ SFC명현고우2μg면역조,초차면역후3주IFN-γ SFC개시하강,다태자격IFN-γ SFC고우단백자격,다태농도위2μg즉가충분자격반점형성;증효을간역묘면역조IFN-γ SFC고우상규을형간염역묘조。결론 초보건립료면역복합형증효을형간염역묘세포면역검측방법병증명기중복성량호,기세포면역수평고우상규을형간염역묘。
Objective To establish the method to detect the cellular immune response of enhanced hepatitis B vaccine and make verification preliminary. Methods Immunized BALB/c mice with enhanced hepatitis B vaccine and detected the IFN-γ spots forming ceils (SFC) of mouse spleen cell by Elispot. Optimized the conditions of the experiment. Cellular immune response between enhanced hepatitis B vaccine and normal hepatitis B vaccine by Elispot were compared. Results IFN-γSFC was higher in 5μ g dose than in 2μg dose after immunization with enhanced hepatitis B vaccine and IFN-γ SFC was declined after immunization 3 weeks ago. IFN-γ SFC was higher in stimulus by peptide than by protein. Compared to normal hepatitis B vaccine, IFN-γ SFC was higher in enhanced hepatitis B vaccine. Conclusion Established the detection method to evaluate the cellular immunity of enhanced hepatitis B vaccine and tested the repeatability.
