目的 探讨臭氧氧化预处理通过诱导热休克蛋白70(HSP70)的合成,保护大鼠肾脏缺血再灌注损伤的作用与机制.方法 建立原位大鼠单侧肾缺血再灌注动物模型,I/R前15 d经直肠吹人氧气和臭氧的混合气体5.0~5.5 ml(臭氧浓度50 mg/L,1 mg/kg体重,每日1次).全自动生化分析仪检测尿素氮(BUN)、肌酐(Cr),比色法测定血清的脂质过氧化产物丙二醛(MDA)、超氧化物歧化酶(SOD).Western blot检测HSP70蛋白的含量;逆转录-聚合酶链反应(RT-PCR)方法检测HSPT0的表达.结果 肾缺血再灌注24 h后,血清中BUN、Cr、MDA明显增高,肾组织内HSP70表达明显增强(P<0.05),经臭氧氧化预处理后,血清中的BUN、Cr、MDA均降低,SOD升高;HSP70表达升高更加明显(P<0.05).结论 臭氧氧化预处理可以诱导大鼠肾缺血再灌注组织中HSP70表达,减轻大鼠肾脏缺血再灌注损伤.

목적 탐토취양양화예처리통과유도열휴극단백70(HSP70)적합성,보호대서신장결혈재관주손상적작용여궤제.방법 건립원위대서단측신결혈재관주동물모형,I/R전15 d경직장취인양기화취양적혼합기체5.0~5.5 ml(취양농도50 mg/L,1 mg/kg체중,매일1차).전자동생화분석의검측뇨소담(BUN)、기항(Cr),비색법측정혈청적지질과양화산물병이철(MDA)、초양화물기화매(SOD).Western blot검측HSP70단백적함량;역전록-취합매련반응(RT-PCR)방법검측HSPT0적표체.결과 신결혈재관주24 h후,혈청중BUN、Cr、MDA명현증고,신조직내HSP70표체명현증강(P<0.05),경취양양화예처리후,혈청중적BUN、Cr、MDA균강저,SOD승고;HSP70표체승고경가명현(P<0.05).결론 취양양화예처리가이유도대서신결혈재관주조직중HSP70표체,감경대서신장결혈재관주손상.
Objective To investigate the protective effect of ozone oxidative preconditioning (OzoneOP)on heat shock protein 70(HSP70)expression in renal ischemia/reperfusion injury(IVR)and to disscuss its protective mechanisms.Methods A rat L/R model was established.the Fenal OzoneOP was induced by rectal insumation 1 mg/kg,15 ozone treatments,one per day,5-5.5 ml at an ozone concentration of 50 mg/L.The parameters of renal function were determined by auto-biochemical analyzer.The concentrations of malondialdehyde(MDA)and superoxide dismutase(SOD)were measured with chromometry.HSP70 was examined by RT-PCR and Western blot.Results Compared with the sham group,the expression of HSP70,the renal funotion and MDA were increased significantly in the rats undergoing L/R procedure.After OzoneOP,the levels of BUN,Cr and MDA were significantly decreased and SOD and HSP70 significantly increased.Conclusion OzoneOP Can increase the HSP70 expression and alleviate kidney renal I/R injury in rats.
