Albendazole and its metabolites%HPLC%Cyprinus carpio%Plasma
[目的] 探索用HPLC测定鲤血浆中ABZ及其代谢物的方法.[方法] 血浆取自体重为(300±30) g的健康鲤,样品经乙酸乙酯2次提取,探索在1个色谱系统下同时测定鲤血浆中阿苯哒唑及其3种代谢物(亚砜、砜和氨基砜)的方法.[结果] 通过考察,确定在测定波长为292 nm,流动相为甲醇-冰乙酸-水时,以1.0 ml/min的流速经C18柱后4种组分的分离效果最佳,保留时间合适,杂质峰少,且峰形良好.在浓度为0.125~32.000 μg/ml的范围内,阿苯哒唑及其3种代谢物平均回收率83.0%~89.2%,日内变异系数2.26%~7.59%,日间变异系数0.45%~2.34%,检测限分别为0.02、0.03、0.05和0.05 μg/ml,定量限分别为0.02、0.03、0.05和0.05 μg/ml.[结论] 建立了利用RP-HPLC同时检测鲤血浆中阿苯哒唑及其3种代谢物的方法.该法操作简单、快速,回收率高,重现性好.

[목적] 탐색용HPLC측정리혈장중ABZ급기대사물적방법.[방법] 혈장취자체중위(300±30) g적건강리,양품경을산을지2차제취,탐색재1개색보계통하동시측정리혈장중아분달서급기3충대사물(아풍、풍화안기풍)적방법.[결과] 통과고찰,학정재측정파장위292 nm,류동상위갑순-빙을산-수시,이1.0 ml/min적류속경C18주후4충조분적분리효과최가,보류시간합괄,잡질봉소,차봉형량호.재농도위0.125~32.000 μg/ml적범위내,아분달서급기3충대사물평균회수솔83.0%~89.2%,일내변이계수2.26%~7.59%,일간변이계수0.45%~2.34%,검측한분별위0.02、0.03、0.05화0.05 μg/ml,정량한분별위0.02、0.03、0.05화0.05 μg/ml.[결론] 건립료이용RP-HPLC동시검측리혈장중아분달서급기3충대사물적방법.해법조작간단、쾌속,회수솔고,중현성호.
[Objective] The aim of research was to explore the method of determining the albendazole and its metabolites in plasma of Cyprinus carpio by HPLC.[Method] The plasma samples were selected from the health C.carpio with weight of (300±30)g and extracted two times by ethyl ecetate so as to explore the method of determining the albendazole and its metabolites (albendazole sulfone, albendazole sulfoxide and albendazole 2-aminosulphone) in plasma of C.carpio under one chromatogram system.[Result] Through investigation, it was confirmed that the 4 components isolated from C18 column with flow velocity of 1.0 ml/min under the detected wavelength of 292 nm and the mobile phase of methanol, water and acetic acid could get the optimum separated effect, with suitable the reserved time, less pmpurity peak and good peak shape.In the range of 0.125-32.000 μg/ml, the recovery rates of albendazole and its metabolites were 83.0%-89.2%, the coefficient of variation ranged in 2.26%-7.59% within one day and 0.45%-2.34% among days and their detection limits were 0.02, 0.03, 0.05 and 0.05 μg/ml resp.and their quantization limits were 0.02, 0.03, 0.05 and 0.05 μg/ml resp.[Conclusion] The method of determining the albendazole and its metabolites in plasma of C.carpio was established by using reversed-phase HPLC, which had characters of simple and rapid operation,high yield and good reproducibility.
