以染料木素为指标,采用紫外分光光度法测定异黄酮的含量,并利用ADS-7大孔树脂分离纯化淡豆豉中异黄酮.结果表明:该树脂分离淡豆豉异黄酮的工艺参数为:上样液浓度0.225 g · mL~(-1),pH值4.5,最大上样量150 mL,以4 BV · h~(-1)的流速吸附,以8倍柱床体积的70%乙醇作为洗脱剂,洗脱流速 4 BV · h~(-1),树脂可重复使用2次.经初步吸附分离,异黄酮得率达40%以上.

Use ultraviolet spectrophotometry to detect the content of soybean isoflavone,with the content of genistein as the index,to investigate the ADS-7 macroporous adsorptive resins for separation and purification of soybean isoflavone in the Semen Sojea Praeparatum. The optimized technological parameters of the ADS-7 macroporous adsorptive resins were as following:loading sample density 0.225 g · mL~(-1),pH 4.5,the maximum sample volume 150 mL,4 BV · h~(-1) flow rate to adsorption and the volume of 70%(v/v)ethanol as 8 BV with desorption-flow 4 BV · h~(-1). With these conditions,more than 40% of soybean isoflavone could be obtained from primary adsorption and purification,and the ADS-7 macroporous adsorptive resins can be used twice.
