目的 在表达绿色荧光蛋白( GFP)裸小鼠脑内建立人脑多形性胶质母细胞瘤(GBM)原位移植模型.方法 将表达GFP的C57/B6鼠与BALB/c nu/nu系裸小鼠杂交筛选出表达GFP的裸小鼠.把人脑GBM组织原位移植于GFP裸小鼠脑内,荧光显微镜下观察移植瘤的生长特性、血管生成及表皮生长因子受体(EGFR)表达.结果 成功培育出表达GFP的荧光裸小鼠;移植瘤中EGFR阳性表达率(78.6±5.2)%,肿瘤侵袭性生长并血管丰富.结论 人脑GBM荧光裸小鼠原位移植模型能模拟人脑原发恶性胶质瘤高侵袭及EGFR高表达的生物学特性.
目的 在錶達綠色熒光蛋白( GFP)裸小鼠腦內建立人腦多形性膠質母細胞瘤(GBM)原位移植模型.方法 將錶達GFP的C57/B6鼠與BALB/c nu/nu繫裸小鼠雜交篩選齣錶達GFP的裸小鼠.把人腦GBM組織原位移植于GFP裸小鼠腦內,熒光顯微鏡下觀察移植瘤的生長特性、血管生成及錶皮生長因子受體(EGFR)錶達.結果 成功培育齣錶達GFP的熒光裸小鼠;移植瘤中EGFR暘性錶達率(78.6±5.2)%,腫瘤侵襲性生長併血管豐富.結論 人腦GBM熒光裸小鼠原位移植模型能模擬人腦原髮噁性膠質瘤高侵襲及EGFR高錶達的生物學特性.

Objective To establish orthotopic xenograft model of human glioblastoma multiforme (GBM) in the brain of nude mice with green fluorescent protein (GFP) expression.Methods Nude mice with ubiquitous GFP expression were obtained by crossing BALB/c nu/nu nude mice with transgenic C57/B6-GFP mice.Then,the fresh tumor tissues of human GBM were orthotopically implanted into the brains of GFP nude mice.The biologic features with growth pattern,angiogenesis and epidermal growth factor receptor (EGFR) expression were determined by fluorescence microscopy.Results GFP nude mice were generated by reciprocally crossing female C57/B6-GFP mice with male BALB/c nu/nu nude mice.The orthotopic xenografts showed invasive behavior,high EGFR expression profile with (78.6 ± 5.2 )% positive rate,and rich angiogenesis under fluorescence background.Conclusion The orthotopic xenografts from human GBM grown in the GFP nude mice shared the same biological features with high EGFR expression and invasiveness with its counterpart in the original tumors.
