目的:探讨系统性红斑狼疮(SLE)患者外周血单个核细胞中核因子(NF)-κB信号通路中IκB激酶(IκK)-α、IκK-β、IκB-α mRNA的定量表达水平,及与SLE病情活动程度的相关性.方法:采集110例SLE患者和50名正常人外周血,抽提总RNA并反转录成cDNA,运用实时定量PCR法在基因测序仪上检测IκK-α、IκK-β、IκB-αmRNA的表达水平.结果:①SLE患者组的IκK-α、IκK-β mRNA定量表达水平显著高于正常对照组(P<0.01);未使用糖皮质激素的初发SLE患者IκK-α、IκK-β mRNA定量表达水平显著高于已使用糖皮质激素的SLE患者(P<0.05);②SLE患者组的IκB-αmRNA定量表达水平显著低于正常对照组(P<0.01);未使用糖皮质激素的初发SLE患者IκB-αmRNA定量表达水平显著低于已使用糖皮质激素的SLE患者(P<0.01);③IκB-α mRNA的表达与IκK-α、IκK-β mRNA表达水平呈负相关(P=0.000);④IκB-α mRNA的表达与SLE疾病活动指数(SLEDAI)积分呈负相关(P<0.01).结论:SLE患者体内NF-κB信号通路处于高活性状态,且与疾病活动程度相关,阻抑NF-κB过度活化可能有利于SLE的治疗.

목적:탐토계통성홍반랑창(SLE)환자외주혈단개핵세포중핵인자(NF)-κB신호통로중IκB격매(IκK)-α、IκK-β、IκB-α mRNA적정량표체수평,급여SLE병정활동정도적상관성.방법:채집110례SLE환자화50명정상인외주혈,추제총RNA병반전록성cDNA,운용실시정량PCR법재기인측서의상검측IκK-α、IκK-β、IκB-αmRNA적표체수평.결과:①SLE환자조적IκK-α、IκK-β mRNA정량표체수평현저고우정상대조조(P<0.01);미사용당피질격소적초발SLE환자IκK-α、IκK-β mRNA정량표체수평현저고우이사용당피질격소적SLE환자(P<0.05);②SLE환자조적IκB-αmRNA정량표체수평현저저우정상대조조(P<0.01);미사용당피질격소적초발SLE환자IκB-αmRNA정량표체수평현저저우이사용당피질격소적SLE환자(P<0.01);③IκB-α mRNA적표체여IκK-α、IκK-β mRNA표체수평정부상관(P=0.000);④IκB-α mRNA적표체여SLE질병활동지수(SLEDAI)적분정부상관(P<0.01).결론:SLE환자체내NF-κB신호통로처우고활성상태,차여질병활동정도상관,조억NF-κB과도활화가능유리우SLE적치료.
Objective: To investigate the mRNA expression levels of IκB kinase (IκK-α)、β IκK-β、IκB-α of NF-κBsignaling path-way in the peripheral blood mononuclear cells of patients with systemic lupus erythematosus (SLE), and to explore the relation of NF-κBsignaling pathway and disease activity of systemic lupus erythematosus. Methods: SYBR green dye I based real-time quantitative PCR method was used to compare the mRNA expression levels of IκK-α IκK-β and IκB-α in the peripheral blood mononuclear cells of SLE patients and healthy controls. Results: (1) IκK-α、 IκK-β mRNA expression levels in SLE patients were significantly higher than those of in normal controls (P< 0.05). IκK-α , IκK-β mRNA expression levels in incipient SLE patients without corticoasteroid therapy were significantly higher than the patients treated with glucocorticoid (P < 0.01). (2) IκB-α mRNA expression level in SLE patients was significantly lower than that of in normal controls (P < 0.01), IκB-α mRNA expression levels in incipient SLE patients with-out corticosteroid therapy were significantly lower than patients treated with glucocorticoid (P < 0.01). (3)IκB-α mRNA expression level in SLE patients correlated negatively with the expression levels of IκK-α and IκK-β RNA (P = 0.000). (4)IκB-α mRNA expres sion level in SLE patients correlated negatively with the SLEDAI score. Conclusion : There is a high expression of NF-κBsignaling pathway in PBMC from SLE patients. The activation of NF-κBsignaling pathway is related to SLE disease activity. Inhibiting the ex-pression of NF-κBsignaling pathway may be beneficial for the treatment of SLE.
