目的 观察化疗前后不同时期胶质瘤组织中毛细血管扩张性共济失调突变基因( ATM)表达变化.方法 利用人胶质瘤U251细胞裸鼠皮下异位移植制作胶质瘤化疗模型,通过顺铂对荷瘤裸鼠进行化疗(每天5 mg/kg,连续腹腔注射4d),建立胶质瘤化疗和化疗后复发模型.利用功能分类基因芯片系统检测化疗前后不同时期胶质瘤组织中ATM的表达,并利用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学法进行验证.结果 ATM在化疗前后不同时期表达出现明显变化,化疗结束时ATM相对表达量较化疗前明显上调(27.96±14.78比1.00±0.00,P<0.01),在化疗后肿瘤消退至最小时ATM表达又明显下调(4.40±3.41,P<0.05),然而在胶质瘤复发时其表达再次明显上调(30.15±7.02,P<0.01),定量RT-PCR和免疫组织化学结果与此一致.结论 化疗前后不同时期胶质瘤组织中ATM的表达呈动态变化并存在时间窗口特征.
目的 觀察化療前後不同時期膠質瘤組織中毛細血管擴張性共濟失調突變基因( ATM)錶達變化.方法 利用人膠質瘤U251細胞裸鼠皮下異位移植製作膠質瘤化療模型,通過順鉑對荷瘤裸鼠進行化療(每天5 mg/kg,連續腹腔註射4d),建立膠質瘤化療和化療後複髮模型.利用功能分類基因芯片繫統檢測化療前後不同時期膠質瘤組織中ATM的錶達,併利用逆轉錄-聚閤酶鏈反應(RT-PCR)和免疫組織化學法進行驗證.結果 ATM在化療前後不同時期錶達齣現明顯變化,化療結束時ATM相對錶達量較化療前明顯上調(27.96±14.78比1.00±0.00,P<0.01),在化療後腫瘤消退至最小時ATM錶達又明顯下調(4.40±3.41,P<0.05),然而在膠質瘤複髮時其錶達再次明顯上調(30.15±7.02,P<0.01),定量RT-PCR和免疫組織化學結果與此一緻.結論 化療前後不同時期膠質瘤組織中ATM的錶達呈動態變化併存在時間窗口特徵.

Objective To study the expression of ataxia-telangiectasia mutated (ATM) in preand post-chemotherapeutic glioma.Methods Ectopicly subcutaneous glioma transplantation models in nude mice were established by injection of human glioma cell line U251 cells.Tumor-bearing nude mice were treated by the intraperitoneal injection of cisplatin [ at the dose of 5 mg/( kg· day) for 4 days continuously ],and chemotherapeutic and recurrent glioma models after chemotherapy were established.The expression of ATM in pre- and post-chemotherapeutic glioma was detected by using oligo toxicology & drug resistance microarrays,reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry.Results The expression of ATM was significantly increased in gliomas after chemotherapy (27.96 ±14.78 vs 1.00 ±0.00,P <0.01 ),significantly decreased when the size of gliomas was smallest (4.40 ±3.41,P < 0.05 ),and increased obviously again when gliomas relapsed ( 30.15 ± 7.02,P < 0.01 ).RT-PCR and immunohistochemistry revealed the similar results.Conclusion The expression of ATM changes ambulatorily in gliomas pre- and post-chemotherapy and there exists a time window.
