目的 探讨大鼠去卵巢后自由基的变化以及胎肾细胞悬液对其干预作用.方法 48只SD大鼠,取8只作为正常对照组,行假手术;其余40只行去卵巢手术,术后随机分为4组.正常对照组及模型对照组给予生理盐水;阳性对照组给予尼尔雌醇;胎肾活细胞悬液组给予活细胞悬液、胎肾死细胞悬液组给予死细胞悬液.药物干预4周后,检测血清超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量、一氧化氮(NO)含量以及一氧化氮合酶(NOS)活力.结果 与正常对照组比较,模型对照组血清SOD、NOS活力以及NO含量均明显下降,MDA含量明显上升(均P<0.01);与模型对照组比较,胎肾活细胞悬液组及胎肾死细胞悬液组SOD活力均有上升(P<0.05,P<0.01);NO含量、NOS活力均明显上升(P<0.05);MDA含量均明显降低(P<0.01);干预3组间两两比较,差异均无统计学意义(P>0.05).结论 去卵巢后大鼠自由基增高,应用胎肾细胞悬液进行干预,能减轻自由基的损伤,增强机体的抗氧化能力.

목적 탐토대서거란소후자유기적변화이급태신세포현액대기간예작용.방법 48지SD대서,취8지작위정상대조조,행가수술;기여40지행거란소수술,술후수궤분위4조.정상대조조급모형대조조급여생리염수;양성대조조급여니이자순;태신활세포현액조급여활세포현액、태신사세포현액조급여사세포현액.약물간예4주후,검측혈청초양화물기화매(SOD)활력、병이철(MDA)함량、일양화담(NO)함량이급일양화담합매(NOS)활력.결과 여정상대조조비교,모형대조조혈청SOD、NOS활력이급NO함량균명현하강,MDA함량명현상승(균P<0.01);여모형대조조비교,태신활세포현액조급태신사세포현액조SOD활력균유상승(P<0.05,P<0.01);NO함량、NOS활력균명현상승(P<0.05);MDA함량균명현강저(P<0.01);간예3조간량량비교,차이균무통계학의의(P>0.05).결론 거란소후대서자유기증고,응용태신세포현액진행간예,능감경자유기적손상,증강궤체적항양화능력.
Objective To observe the changes of free radicals in rats after ovariectomy and the intervention effect of fetal renal cell suspension(FRCS)on them.Methods Totally 48 Sprague-Dawley rats,eight in the normal control group were sham-operated and treated with saline; the other 40 were ovariectomized and randomly divided into four groups: the model control group(A)administered with normal saline,the positive control group(B)administered with nilestriol,the two testing groups(C and D)administered respectively with living and dead FRCS.The administration was beginning from 12 weeks after operation and lasted for four weeks.Levels of serum superoxide dismutase(SOD)and nitric monoxide synthase(NOS)activity,malondialdehyde(MDA)and nitric oxide(NO)contents were measured at the terminal of the study.Results Compared with the shamoperated group,levels of SOD,NOS and NO in Group A were significantly lower,while level of MDA was significantly higher(P<0.01).Compared with Group A,all above-mentioned abnormalities of indices were inversely changed in the three intervened groups significantly(P<0.05 or P<0.01),but showed insignificant difference in the paired comparisons of the three groups(P>0.05).Conclusion High free radical condition is surely present in ovariectomized rats,FRCS can lessen the injury of free radicals and enhance the oxidation antagonizing capacity of the organism.
