目的 观察原发性胆汁性肝硬化(PBC)患者外周血T细胞中微小RNA( microRNA,miRNA)表达谱的变化.方法 选取20例PBC患者,收集同期20份健康人外周血作对照.应用芯片技术筛选PBC患者外周血T细胞中表达差异的miRNA,并用实时定量PCR进行验证.结果 与健康对照组相比,PBC患者外周血T细胞中有25个miRNA表达失调,其中有23个下调,仅有2个上调(均P<0.05).定量PCR显示,miR-346、miR-17-5p、miR-20a和miR-let-7b表达显著低于健康对照,而miR-451和miR-129表达显著高于健康对照,与芯片结果一致.结论 PBC患者外周血T细胞miRNA表达与健康对照比较存在差异,这些异常表达的miRNA可能参与PBC的发病.

목적 관찰원발성담즙성간경화(PBC)환자외주혈T세포중미소RNA( microRNA,miRNA)표체보적변화.방법 선취20례PBC환자,수집동기20빈건강인외주혈작대조.응용심편기술사선PBC환자외주혈T세포중표체차이적miRNA,병용실시정량PCR진행험증.결과 여건강대조조상비,PBC환자외주혈T세포중유25개miRNA표체실조,기중유23개하조,부유2개상조(균P<0.05).정량PCR현시,miR-346、miR-17-5p、miR-20a화miR-let-7b표체현저저우건강대조,이miR-451화miR-129표체현저고우건강대조,여심편결과일치.결론 PBC환자외주혈T세포miRNA표체여건강대조비교존재차이,저사이상표체적miRNA가능삼여PBC적발병.
Objective To explore the expression pattenn of microRNA (miRNA) in T cells of peripheral blood mononuclear cell (PBMC) from patients with primary biliary cirrhosis (PBC).Methods The expression profile of miRNA in T cells of PBMC was determined by microarray assay and validated by real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR).Results In comparison with the healthy controls,23 miRNA were down-regulated and 2 miRNA had a higher expression ( all P <0.05). As revealed by qRT-PCR,the expressions of miR-346,miR-17-5p,miR-20a and miR-let-7b decreased obviously while miR-451 and miR-129 became up-regulated.The results were in agreement with those of microarray. Conclusions The PBC patients and healthy controls have significantly different expression profiles of microRNA in T cells of PBMC. The differential expression of microRNA may be involved in the pathogenesis of PBC.
