TWIST是胚胎发育过程中的重要转录调控因子,最近发现它在肿瘤转移的起始阶段使癌细胞发生EMT现象,即细胞从上皮极性表型向间质成纤维细胞表型的形态学上的转变,同时伴有上皮细胞分子标记的抑制和间质细胞分子标记的强烈表达,从而促进细胞的运动和迁移,使肿瘤呈现恶性表型,导致患者预后不良.目前已经证实有多种细胞信号通路和细胞内因子参与了TWIST的表达和激活,它们对TWIST功能起重要的调控作用.本文主要就目前TWIST的结构、功能及在肿瘤中的作用作一综述.
TWIST是胚胎髮育過程中的重要轉錄調控因子,最近髮現它在腫瘤轉移的起始階段使癌細胞髮生EMT現象,即細胞從上皮極性錶型嚮間質成纖維細胞錶型的形態學上的轉變,同時伴有上皮細胞分子標記的抑製和間質細胞分子標記的彊烈錶達,從而促進細胞的運動和遷移,使腫瘤呈現噁性錶型,導緻患者預後不良.目前已經證實有多種細胞信號通路和細胞內因子參與瞭TWIST的錶達和激活,它們對TWIST功能起重要的調控作用.本文主要就目前TWIST的結構、功能及在腫瘤中的作用作一綜述.

Objective As a important transcriptional factor during the embryonic development process,recently TWIST has been found to promote the EMT phenomenon in the initiation step of tumor metastasis.EMT is a morphological switch of cells from the epithelial polarized phenotype to the mesenchymal fibroblastoid phenotype,accompanying obvious lost of epithelial marker and strong gain of mesenchymal marker in cells which can result in the tumor manifests malignant phenotype and poor prognosis.At present,many of signal transduction pathways and cellular factors have been reported to involve in the expression and activation of TWIST.In this review,we will comprehensively discuss the progress on the structure,function and the role in tumor of TWIST.
