目的 观察特菲那丁对豚鼠QT间期(QTc)的影响及其对快速室性心律失常的防治作用.方法 (1)腹腔注射特非那丁后观察其对豚鼠QTc的影响.(2)腹腔汴射特非那丁后,观察氯化钡诱发快速室性心律失常的难易程度.同时采用胺碘酮作为阳性对照.结果 (1)不同浓度的特非那丁在腹腔注射后呈剂量依赖性延长QTc,与生理盐水、DMSO组及给药前比较差异有统计学意义(P<0.05).(2)特非那丁可明显缩短3种剂量氯化钡所致室性心动过速(审速)持续的时间.与生理盐水组比较差异有统计学意义(P<0.05),且特非那丁对氯化钡致室速的防治作用不低于胺碘酮.结论 (1)特非那丁可剂量依赖性地延长QTc间期.(2)特非那丁可显著缩短窒速持续的时间

Objective To evaluate the effects of terfenadine on QTc interval and on an experimental ventricular arrhythmia in guinea pigs.Methods (1)The effect of terfenadine on the QTc interval in guineapigs wag evaluated by comparison of multiple ECGs.(2)To induce an experimental arrhythmia,an intraperitoneal injection of BaCh was administered.Saline and amiodarone were administered as intraperitoneal injections in separate guinea pigs,which served as controls.Results (1)In comparison with normal saline and amiodalone,terfenadine caused prolonged QTc interval in the guinea pig that was dose-dependent.(2)In the BaCl2 induced experimental arrhythmia model,terfenadine caused a significant,dose-dependent shortening of the duration of ventricular arrhythmia in comparison with normal saline control(P<0.05).Conclusion The results suggest terfenadine appears to prolong the QTc interval and decrease ventricular arrhythmia duration in a dosedependent manner in guinea pigs.
