采用改良甲苯胺蓝(MTB)、阿利新蓝-沙黄(AB-S)、甲基绿-派洛宁(MG-P)、天青Ⅱ-伊红-瑞特、硫堇及PAS-HE等6种组织化学染色方法,对虎纹蛙消化道肥大细胞(MC)的组化性质进行了研究.结果表明:虎纹蛙消化道中食管、胃贲门、胃体、胃幽门、空肠、十二指肠、回肠、直肠等部位均有MC分布;MC呈长梭形、圆形、卵圆形、不规则形,主要存在于黏膜固有层或黏膜下层内,有沿血管和腺泡周围分布的趋向.在6种染色法中,用MTB染色效果最好,MC易定位,组织结构清晰可辨,胞质呈紫红色;其次为AB-S染色,而MG-P及天青Ⅱ-伊红-瑞特的染色效果一般,硫堇染色效果差,PAS-HE染色则不着色.

Histochemical property of mast cells (MCs) in the digestive tract of Hoplobatrachus rugulosus was studied by using six staining methods, such as modified toluidine blue (MTB), alcian blue-shahuang (AB-S), methyl green-pyronin (MG-P), azure Ⅱ-eosin-wright, thionine and PAS-HE and so on. The results showed that there were MCs in the esophagus, stomachus cardiacus, gastric body, stomachus pyloricus, jejumum, duodenum, ileum and rectum of digestive tract of H.rugulosus, and MCs had long fusiform, round, ovoid and irregular shapes. MCs mainly existed in the lamina propria or lower layer of the mucosa, and had trend of around blood vessel and acinar. In the six staining methods, MTB staining had the best effect, MCs were easy to be located, the tissue structure could be seen clearly, and cell plasma was purple red. AB-S staining had better effect, MG-P and azure Ⅱ-eosin-wright staining had common effect, thionine staining had bad effect, and PAS-HE staining had no coloring.
