目的 研究小窝蛋白-1(caveolin-1)在人胶质瘤中的表达,探讨其在胶质瘤的发生、发展及细胞增殖中的作用.方法 应用免疫组化SP法检测80例胶质瘤和9例正常脑组织中caveolin-1与增殖细胞核抗原(PCNA)的表达情况.结果 caveolin-1在正常脑组织中全部阳性表达.80例胶质瘤中39例caveolin-1呈阳性表达,其在低级别胶质瘤中的阳性表达率高于高级别胶质瘤(P<0.05);PCNA在正常脑组织中未见表达,80例胶质瘤中55例呈阳性表达,其在高级别胶质瘤中的阳性表达率高于低级别胶质瘤(P<0.05).caveolin-1与PCNA在胶质瘤中的表达呈负相关性(r=-0.368,P<0.01).结论 caveolin-1在胶质瘤中的表达对肿瘤细胞增殖起抑制作用,为胶质瘤的基因治疗提供了一个新的靶点.PCNA可作为反映胶质瘤细胞增殖、恶性程度及预后的指标之一.

Objective To explore the expression of caveolin-1 in human gliomas and its effects on the development and cell proliferation of gliomas.Methods The expression and distribution of caveolin-1 and proliferating cell nuclear antigen (PCNA) were determined by immunohistochemical technique in 80 cases of glioma specimens and 9 normal brain tissues.Results Caveolin-1 was expressed positively in all normal brain tissues,and in 39 of 80 cases of glioma specimens,higher in low grade glioma specimens than in high (P<0.05).PCNA was not expressed in normal brain tissues,but expressed in 55 cases of glioma specimens,higher in high grade glioma than in low (P<0.05).There was negative correlation between expressions of caveolin-1 and PCNA in glioma(r=-0.368,P<0.01).Conclusion Caveolin-1 in glioma has depressant effects on tumor cell proliferation,providing a new target for the glioma gene therapy.PCNA can be used as one of prognostic indicators reflecting glioma cell proliferation,malignant degree and its prognosis.
