目的 探讨血管紧张素转换酶(angiotensin coverting enzyme,ACE)基因插/缺失(I/D)多态性与广西巴马长寿的关系.方法 采用聚合酶链反应(PCR)、单链构象多态性分析(SSCP)与直接测序技术,研究百岁组(≥100岁)27例、90岁组(90~99岁)56例、老年组(60~89岁)114例老人的ACE基因I/D多态性,以79例60~70岁自然死亡人群为对照组.结果 在ACE基因I/D多态基因型中,百岁组L/I、L/D、D/D型频率分别为37.04%、33.33%和29.63%,I、D等位基因频率分别为53.70和46.30%.百岁组D/D基因型频率和D型等位基因频率高于老年组(P<0.05)且明显高于对照组(P<0.01),但与90岁组比较,无论是I/I、I/D或D/D基因型,所占百分率均比较接近,差异无显著性(P>O.05).D/D基因型经SSCP证实全部相同,无差别.直接测序结果再次验证ACE(I/D)多态性判断准确.结论 ACE基因I/D多态性与长寿密切相关.长寿受多种因素影响.

Objective To study the distribution of I/D polymorphisms of angiotensin coverting enzyme(ACE)gene and its relationship to longevity of Bama.Methods Polymerize chain reaction,single strand conformation polymorphism,and direct sequencing technique were used to test I/D polymorphisms of ACE gene in 27 centenarians,56persons aged 90~99,114 persons aged 60~89 and 79 cases of natural death aged 60~70 were used as controls.Resuits The frequencies of ACE genotypes I/I,I/D and D/D were 37.04%,33.33%,and 29.63% respectively in the centenarian group.The frequency rates of I and D alleles were 53.70% and 46.30%.The frequency rate of D/D genotype of the centenarian group was higher than that of the group aged 60~89(X~2=4.339,P<0.05),significantly higher than that of the group aged 60~70(X~2=7.145,P<0.01).However,it is not significantly different from that of the group aged 90~99(P>0.05).The frequency rate of D allele of the group of centenarian was higher than that of the group aged 60~70(X~2=4.466,P<0.05).The frequency rates of genotype D/D and D alleles were significantly higher in the centenarian group than that in the controls(both P<0.01).There is no difference between D/D genotype by single strand conformation polymorphism.Direct sequencing results reconfirmed the veracity of I/D polymorphisms of ACE gene.Conclusion ACE gene polymorphism is closely correlated with life span.In addition,longevity is the interaction of multiple factors.
