从材料预处理方法、消毒试剂、选材、培养基及季节等方面,研究尾巨桉家系外植体诱导的相关技术。结果表明:用饱和洗衣粉水泡洗外植体5分钟后再用毛刷清洗其表面污物的预处理方法效果最佳,其污染率较低,为38%,而萌芽率较高,达62%;消毒试剂使用酒精加氯化汞消毒1次效果最好,污染与灼伤较少,且出芽率高,达68%;取材部位以伐倒后的萌芽诱导效果最好,有效出芽率高达32%;诱导培养基的激素浓度以1.2nag·L^-1BA,0.6nag·L^-1NAA搭配较好,增殖倍数适宜,有2~3个芽,且长势较好;外植体诱导全年均可进行,春季萌芽率高但污染率也高,冬季污染率低,但萌芽率低,生长势弱。

Related techniques including pretreatments methods, disinfecting reagents, materials, culture medium and seasons for inducing explants of E. urophylla x E. grandis families were carried out. The results showed that the best pretreatment method was to soak explants by saturated detergent for 5 minutes and then to clear the surface pollutants by brash,which could get the lowest polluted rate of 38% and the highest germinating rate of 62% ; the best disinfecting reagent was alcohol with I-IgC12 used to disinfect one time, which could have lower polluted and burned rate and the highest germination rote of 68% ; the best materials for induehag was sprouts after cutting trees, which had good germinating rate of 32% ; the best hormone concentration of culture medium was 1.2 nag ·L^-1 BA with 0.6 mg · L^-1 NAA which could get appropriate propagation results and grow well with 2- 3 buds. Explant induction could be done in any season, high germinating and polluted rates were in spring, and the opposite in winter.
