目的:观察人胚胎嗅鞘细胞 (olfactory ensheathing cells, OECs)移植对成鼠脊髓轴突再生的影响. 方法将分离、纯化、培养 2周的人胚胎 OECs,移植于 10只成年 SD大鼠半横断胸髓( T10)的两端;对照组 10只同样方法给予等量的 DMEM培养液. 6周后组织切片,行 HE染色、嗜银染色以及髓鞘碱性蛋白( MBP)和抗神经生长因子受体抗体( NGFR)免疫组化检查. 结果 OECs移植组 6周后脊髓大体标本显示初步愈合现象;组织学观察, OECs呈 NGFR阳性表达,显示 OECs仍然存活,且与宿主组织整和良好;部分再生轴突长入断端间组织,呈 MBP阳性表达. 结论人胚胎 OECs对成鼠胸髓损伤后的轴突再生有促进作用.
目的:觀察人胚胎嗅鞘細胞 (olfactory ensheathing cells, OECs)移植對成鼠脊髓軸突再生的影響. 方法將分離、純化、培養 2週的人胚胎 OECs,移植于 10隻成年 SD大鼠半橫斷胸髓( T10)的兩耑;對照組 10隻同樣方法給予等量的 DMEM培養液. 6週後組織切片,行 HE染色、嗜銀染色以及髓鞘堿性蛋白( MBP)和抗神經生長因子受體抗體( NGFR)免疫組化檢查. 結果 OECs移植組 6週後脊髓大體標本顯示初步愈閤現象;組織學觀察, OECs呈 NGFR暘性錶達,顯示 OECs仍然存活,且與宿主組織整和良好;部分再生軸突長入斷耑間組織,呈 MBP暘性錶達. 結論人胚胎 OECs對成鼠胸髓損傷後的軸突再生有促進作用.
목적:관찰인배태후초세포 (olfactory ensheathing cells, OECs)이식대성서척수축돌재생적영향. 방법장분리、순화、배양 2주적인배태 OECs,이식우 10지성년 SD대서반횡단흉수( T10)적량단;대조조 10지동양방법급여등량적 DMEM배양액. 6주후조직절편,행 HE염색、기은염색이급수초감성단백( MBP)화항신경생장인자수체항체( NGFR)면역조화검사. 결과 OECs이식조 6주후척수대체표본현시초보유합현상;조직학관찰, OECs정 NGFR양성표체,현시 OECs잉연존활,차여숙주조직정화량호;부분재생축돌장입단단간조직,정 MBP양성표체. 결론인배태 OECs대성서흉수손상후적축돌재생유촉진작용.
AIM:To observe the effect of transplantation of human embryonic OECs on adult rats' semitransected spinal cord. METHODS: OECs,separated from human embryonic olfactory bulb and purified by culture in vivo for 2 weeks,were transplanted into SD rats'(n=10) semitransected spinal cord at T10 level,while the vehicle solution (DMEM) was injected in the controls(n=10).Samples were procured at 6 week after transplantation and studied by nerve silver staining and myelin basic protein(MBP) histochemical staining and nerve growth factor receptor(NGFR) immunohistochemical staining. RESULTS:The injured spinal cords displayed initial healing in OECs transplanted rats 6 weeks post-transplantation morphologically.Histological observation showed that the transplanted OECs integrated into the host tissues in well arrangement and the regenerative axons grew into the distal end of the semitransected spinal cords.Both of OECs and regenerative axons positively expressed MBP and NGFR.In control group, the ends of the impaired spinal cords exhibited liquefied necrosis and axonal degeneration and no regnnerative axons were found. CONCLUSION:OECs transplantation could protect the impaired spinal cord and promote the regeneration of axons.
