目的探讨IFN-γ抑制人成纤维细胞内弓形虫增殖的机制. 方法用反相高效液相色谱法测吲哚胺2,3过氧化酶(IDO)活性;用3H-Uracil掺入法测弓形虫增殖;用反转录聚合酶链式反应测IDO mRNA的表达. 结果 IFN-γ能抑制人成纤维细胞内弓形虫增殖,培养上清液中检测到色氨酸被降解,生成了犬尿氨酸;培养液中加入足量色氨酸后,IFN-γ对弓形虫的抑制作用被部分逆转;用IDO特异引物扩增出一条662 bp DNA条带. 结论 IFN-γ抑制人成纤维细胞内弓形虫增殖是通过诱导细胞表达IDO活性降解色氨酸机制实现的.

Objective To explore the mechanism of IFN-γ to inhibit the proliferation of Toxoplasma gondii in human fibroblasts. Methods Indoleamine 2,3-dioxygenase(IDO) activity was determined by reverse-phase high performance liquid chromatography. The extent of the inhibition of the growth of T. gondii was measured by the incorporation of 3H-uracil. Expression of IDO mRNA was detected by RT-PCR. Results IFN-γ could induce human fibroblasts to express IDO activity and resist the replication of T. gondii. Depletion of tryptophan and accumulation of kynurenine in culture supernatant were found. A 662 bp DNA band standed for IDO mRNA was detected. Conclusion IFN-γ inhibits the proliferation of T. gondii in human fibroblasts by inducing IDO to degradate tryptophan.
