以杂交一代品种雪山椰菜花和自交系SZ-70为供试材料,研究了不同平阳霉素处理浓度对种子萌发的影响、不同激素种类和浓度对不定芽诱导和增殖培养的效果以及不同平阳霉素处理浓度对离体培养茎尖存活的影响.结果表明:以6-BA 2 mg/L和NAA 0.25 mg/L诱导茎尖产生不定芽的效果优于子叶和下胚轴;6-BA0.2 mg/L+NAA 0.02 mg/L+MET 0.2 mg/L对不定芽的增殖效果最佳;平阳霉素处理对种子的萌发率影响不明显,处理浓度越高,种子萌发后真叶抽出率越低;随着平阳霉素处理浓度的提高,茎尖存活率下降,且异常苗和玻璃苗增加.平阳霉素80 mg/L、30 min的处理对茎尖的诱变效果较为适合.

The F_1 hybrid Xueshan Yecaihua(SN)and the lnbred line SZ-70 bred by the Horticultural Research Institute were used as test materials,and the effects of Pingyangmicin and hormone treatments were studied on the seed germination,adventitious bud induction and propagation,and in vitro culture of stem apexes of B.oleracea vat.botrytis.The results showed that the stem apexes of sterilized seedlings were better than the cotyledons and hypocotyls of them in inducing adventitious buds with 2 mg/L 6-BA and 0.25 mg/L NAA.The medium added with 0.2 mg/L 6-BA,0.02 mg/L NAA and 0.2 mg/L MET had the best effect on the adventitious bud propagation.Pingyangmicin treatments had not an obvious effect on the seed germination,but increasing Pingyangmicin concentration could decrease the true leaf germination rate and the survival rate of the stem apexes and increase the abnormal and vitreous shoots.The treatment with 80 mg/L Pingyangmicin for 30 min was the median lethal dose of the stem apexes.
