目的研究急性白血病患儿反复应用重组粒细胞集落刺激因子(rhG-CsF)的药代动力学及其与临床的关系。方法应用酶联免疫吸附测定(ELIsA)方法检测46例白血病患儿的血清G-CSF浓度。结果获得29例患儿用rgG-CSF后的标本,其血清G-CSF浓度为(5.335±5.597)μg/L。反复应用rhG-CSF的患儿在下次用药前血清G-CSF水平恢复正常;少数未应用rgG-CSF患儿可检测到较高浓度的G-CsF。血清G-CSF浓度与粒细胞绝对计数值呈负相关(r=-0.8746,P<0.01)。结论 rhG-CSF的药代动力学中存在有粒细胞介导的清除机制,反复应用无药物蓄积作用。

목적연구급성백혈병환인반복응용중조립세포집락자격인자(rhG-CsF)적약대동역학급기여림상적관계。방법응용매련면역흡부측정(ELIsA)방법검측46례백혈병환인적혈청G-CSF농도。결과획득29례환인용rgG-CSF후적표본,기혈청G-CSF농도위(5.335±5.597)μg/L。반복응용rhG-CSF적환인재하차용약전혈청G-CSF수평회복정상;소수미응용rgG-CSF환인가검측도교고농도적G-CsF。혈청G-CSF농도여립세포절대계수치정부상관(r=-0.8746,P<0.01)。결론 rhG-CSF적약대동역학중존재유립세포개도적청제궤제,반복응용무약물축적작용。
Objective To investigate the pharmacokinetic progerty after repeated administration of recombinant human granulocyte colony stimulating factor( rhG-CSF) in children with acute leukemia and its clinical significance. Methods . Serum G-CSF concentrations were measured in 46 acute leukemic children by enzyme linked immunosorbent assay. Results The serum G-CSF concentration was(5. 335 ±5. 597 ) μg/L in 29 children during rhG-CSF treatment . In children received repeted treatment of rhG-CSF,serum G-CSF levels resumed to normal before next administration. Only few cildren (2/8)who didnf receive any rhG-CSF treatment had higher rhG-CSF concentrations;Tbe serum G-CSF concentrations correlated negatively with absolute neutrophil counts ( r = - 0. 8746, P < 0. 01 ). Conclusion There is a granulocyte - madiating clearing mechanism in rhG-CSF pharmacokinetics, and repeated administration of rhG-CSF was safe without accumuting effect.
