本文建立了一种用高效液相色谱法同时检测饲料中雌二醇和甲基睾丸酮的新方法。饲料样品经乙酸乙酯提取,采用液液萃取法净化浓缩,经液相色谱分离后用二极管阵列器检测。在0.5~10.0 mg/L浓度范围内,两种激素呈良好的线性关系,相关系数均大于0.9997。在添加水平0.1~2.0 mg/kg范围内,雌二醇的平均回收率为71%~86%,甲基睾丸酮的平均回收率为76%~91%,相对标准偏差均小于9.6%。雌二醇、甲基睾丸酮最低检出限(S/N=3)均为0.03 mg/kg,定量限(S/N=10)为0.1 mg/kg。该方法灵敏度高,测定结果准确,适用于饲料中雌二醇和甲基睾丸酮的同时检测。

A method for the simultaneous determination of estradiol and methyltestosterone in animal feeds by high performance liquid chromatography(HPLC) was developed.The target compounds were first extracted with ethyl acetate and then purified by following liquid-liquid extraction processes.The residues were determined by HPLC with a diode array detector.The standard curves of estradiol and methyltestosterone showed good linearity in the range of 0.5~10.0mg/L,and the correlation coefficients(r) were higher than 0.9997.At the spiked levels range from 0.1 to 2.0 mg/kg,the average recoveries of estradiol and methyltestosterone were 71%-86% and 76%-91%,respectively.The relative standard deviations(RSDs) were both less than 9.6%.The detection limits(LOD,S/N=3) of estradiol and methyltestosterone were both 0.03mg/kg and the quantification limits(LOQ,S/N=10) were both 0.1mg/kg.This method is sensitive,accurate and suitable for the simultaneous determination of estradiol and methyltestosterone in animal feeds.
