目的 观察鞣酸(tannic acid,TA)对链脲佐菌素(STZ)诱发糖尿病(DM)大鼠血糖及肾脏功能和形态改变的影响.方法 用STZ诱发大鼠DM模型后,随机分为DM模型组、氨基胍(AG)组和TA低高剂量组,另设正常对照组.AG组按40 mg·kg~(-1)·d~(-1)给予;TA低高剂量组分别按20及30 mg·kg~(-1)·d~(-1)给予;正常对照组和模型组每天给予等体积生理盐水,给药方式均为腹腔注射,共10 w.观察给药期间及10 w后大鼠的一般状况,检测各组大鼠血糖、血肌酐(Cr)、血尿素氮(BUN)、尿酸(UA)、24 h尿蛋白及观察肾脏形态学变化.结果 TA可改善DM大鼠基本状况,与模型组比较,TA组血糖、血清Cr、BUN和尿Cr、尿蛋白均明显降低,光镜和电镜下观察显示,模型组大鼠较正常对照组大鼠肾小球明显增大,细胞数目增多,基底膜节段性增厚,部分足突融合.TA组上述异常均明显减轻.结论 TA可明显降低尿蛋白排泄,改善DM大鼠肾脏功能和形态学异常,对DM大鼠具有肾脏保护作用.

목적 관찰유산(tannic acid,TA)대련뇨좌균소(STZ)유발당뇨병(DM)대서혈당급신장공능화형태개변적영향.방법 용STZ유발대서DM모형후,수궤분위DM모형조、안기고(AG)조화TA저고제량조,령설정상대조조.AG조안40 mg·kg~(-1)·d~(-1)급여;TA저고제량조분별안20급30 mg·kg~(-1)·d~(-1)급여;정상대조조화모형조매천급여등체적생리염수,급약방식균위복강주사,공10 w.관찰급약기간급10 w후대서적일반상황,검측각조대서혈당、혈기항(Cr)、혈뇨소담(BUN)、뇨산(UA)、24 h뇨단백급관찰신장형태학변화.결과 TA가개선DM대서기본상황,여모형조비교,TA조혈당、혈청Cr、BUN화뇨Cr、뇨단백균명현강저,광경화전경하관찰현시,모형조대서교정상대조조대서신소구명현증대,세포수목증다,기저막절단성증후,부분족돌융합.TA조상술이상균명현감경.결론 TA가명현강저뇨단백배설,개선DM대서신장공능화형태학이상,대DM대서구유신장보호작용.
Objective To investigate the effects of tannic acid (TA) on the renal pathological functional and morphological changes in streptozotocin-induced diabetic rats.Methods Diabetic rats induced by STZ were randomly divided into diabetes,aminoguanidine (AG),low and high dosages of TA groups and daily injected I.p.with physiological saline,AG (40 mg·kg~(-1)·d~(-1)) and TA (20 and 30 mg·kg~(-1)·d~(-1)),respectively,for 10 w.Healthy rats were served as normal control.General situation was observed during and after 10 weeks' treatment,and blood glucose,Cr,BUN,UA,urinary protein were detected.Meanwhile,the morphological changes of kidney were observed by light and electron microscope.Results Treatment of TA improved the general conditions of diabetic rats.Compared with the model group,blood glucose,Cr,BUN and urinary protein were decreased significantly.The conventional and ultrastructure alterations in TA group including volume enlargement of kidney glomerulus,mesangial cells increase and mesangial expansion and thickening of glomerular basement membrane (GBM),were lighter than those in DM group.Conclusions TA could markedly reduce the pathological changes of kidney,inhibit the kidney hypertrophy and reduce the proteinuria of diabetic rats,could effectively protect the function of kidney in diabetic rats.
