目的 观察乳酸.羟基乙酸-L-赖氨酸(RGD)多肽接枝聚复合FK506和神经生长因子(NGF)缓释膜桥接大鼠坐骨神经5 mm缺损促进神经再生的效果.方法 将24只雄性Wistar大鼠随机分成三组,切断左侧坐骨神经形成5 mm缺损,分别以RGD多肽接枝聚复合FK506和NGF缓释膜(A组)、RGD多肽接枝聚复合FK506缓释膜(B组)和RGD多肽接枝聚膜(C组)桥接缺损;A组膜FK506与NGF的含量分别为0.48 mg和2μg,B组膜FK506的含量为0.72 mg,C组为对照组.术后3个月,采用大体观察、神经电生理、小腿三头肌湿重恢复率、组织学观察和图像分析等方法规查神经再生情况. 结果术后3个月,膜外形完整、质脆;各组再生神经均通过了缺损,且A组的再生神经各项检测指标明显优于B组和C组.结论 RGD多肽接枝聚复合FK506和NGF缓释膜能够有效地促进损伤的大鼠坐骨神经再生,且FK506和NGF联合应用,可减少FK506的用量.

Objective To observe the effect of sustained-release diaphragm of the RGD peptide grafted poly[LA-(Glc-Lys)], FKS06 and nerve growth factor(NGF) on bridging a 5 mm gap of sciatic nerve in rats. Methods Twenty-four aduh male Wistar rats were randomly divided into 3 even groups. Defects of 5 mm long were created at the left sciatic nerve in all the rats, and bridged respectively with the sus-tained-release diaphragm of RGD peptide grafted poly[LA-(Glc-Lys)], FKS06 and NGF (Group A), the sustained-release diaphragm of RGD peptide grafted poly[LA-(Glc-Lys)] and FKS06 (Group B) and the di-aphragm of RGD peptide grafted poly[LA-(GIc-Lys)] (Group C). FKS06 and NGF were O. 48 mg and 2 μg respectively in Group A, and FKS06 was 0.72 mg in Group B and Group C as a control. Three months postoperatively, the results were evaluated by gross observing, eleetrophysiology, weighing of gastrocnemius muscles, histological changes and morphometric analysis. Results Three months postoperatively, all the diaphragms were crisp and remained intact in appearance, and the regenerative nerve in the 3 groups had bridged the defects. Group A had better results than Groups B and C in all the indexes examined. Con-clusions The sustained-release diaphragm of RGD peptide grafted pely[LA-(Gle-Lys)], FK506 and NGF can efficiently promote the regeneration of injured sciatic nerve in rats, and reduce the use of FK506 when FK506 and NGF are used together.
