目的:观察神经生长因子(NGF)及其受体(TrkANGFR和p75NTR)在肝细胞中的表达,探讨外源性神经生长因子β(NGF-β)对肝细胞的生物学作用.方法:体外培养L02肝细胞,免疫细胞化学和荧光定量PCR法分别检测NGF、TrkANGFR和p75NTR在L02细胞中的表达.XTT法检测外源性NGF-β、anti-NGF、anti-TrkANGFR和anti-p75NTR对L02细胞增殖的作用,流式细胞术检测外源性NGF-β对L02细胞凋亡和细胞周期的影响.结果:L02细胞表达NGF及其受体TrkANGFR、p75NTR,NGF主要位于细胞质和细胞核,TrkANGFR和p75NTR位于细胞质和细胞膜;外源性NGF-β上调L02细胞表达NGF和TrkANGFR.低剂量外源性NGF-β(12.5~200 μg/L) 通过调控L02细胞的S期,促进细胞增殖,抗细胞凋亡,高剂量(>400 μg/L)NGF-β不能促进L02细胞增殖;anti-NGF和anti-TrkANGFR抑制NGF-β诱导的L02细胞增殖,anti-p75NTR并不影响NGF-β诱导的L02细胞增殖.结论:L02细胞表达NGF及其受体TrkANGFR、p75NTR;合适剂量外源性NGF-β可能通过NGF/TrkANGFR信号途径促进L02细胞增殖.

목적:관찰신경생장인자(NGF)급기수체(TrkANGFR화p75NTR)재간세포중적표체,탐토외원성신경생장인자β(NGF-β)대간세포적생물학작용.방법:체외배양L02간세포,면역세포화학화형광정량PCR법분별검측NGF、TrkANGFR화p75NTR재L02세포중적표체.XTT법검측외원성NGF-β、anti-NGF、anti-TrkANGFR화anti-p75NTR대L02세포증식적작용,류식세포술검측외원성NGF-β대L02세포조망화세포주기적영향.결과:L02세포표체NGF급기수체TrkANGFR、p75NTR,NGF주요위우세포질화세포핵,TrkANGFR화p75NTR위우세포질화세포막;외원성NGF-β상조L02세포표체NGF화TrkANGFR.저제량외원성NGF-β(12.5~200 μg/L) 통과조공L02세포적S기,촉진세포증식,항세포조망,고제량(>400 μg/L)NGF-β불능촉진L02세포증식;anti-NGF화anti-TrkANGFR억제NGF-β유도적L02세포증식,anti-p75NTR병불영향NGF-β유도적L02세포증식.결론:L02세포표체NGF급기수체TrkANGFR、p75NTR;합괄제량외원성NGF-β가능통과NGF/TrkANGFR신호도경촉진L02세포증식.
