目的 观察大蒜油对对乙酰氨基酚(AP)所致小鼠急性肝损伤是否有保护作用.方法 将75只小鼠随机分为阴性对照组、模型组以及大蒜油低、中、高三个剂量组(分别为25、50、100 mg/kg),给药后2 h除阴性对照组其他各组小鼠均灌胃AP 240 mg/kg;24 h后小鼠摘眼球取血,离心,测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)的活性,同时比较各组小鼠的肝脏系数和肝脏病理组织学变化.结果 与模型组相比,大蒜油低、中、高剂量组的肝脏系数明显减小,血清ALT、AST活性显著降低(P<0.01),同时肝脏组织病理学检查发现模型组肝细胞发生气球样变,甚至坏死,炎细胞浸润,而大蒜油组无明显病理学改变.结论 大蒜油对AP所致小鼠急性肝损伤有明显的保护作用.

목적 관찰대산유대대을선안기분(AP)소치소서급성간손상시부유보호작용.방법 장75지소서수궤분위음성대조조、모형조이급대산유저、중、고삼개제량조(분별위25、50、100 mg/kg),급약후2 h제음성대조조기타각조소서균관위AP 240 mg/kg;24 h후소서적안구취혈,리심,측정혈청중병안산안기전이매(ALT)、천문동안산안기전이매(AST)적활성,동시비교각조소서적간장계수화간장병리조직학변화.결과 여모형조상비,대산유저、중、고제량조적간장계수명현감소,혈청ALT、AST활성현저강저(P<0.01),동시간장조직병이학검사발현모형조간세포발생기구양변,심지배사,염세포침윤,이대산유조무명현병이학개변.결론 대산유대AP소치소서급성간손상유명현적보호작용.
Objective To study the pretective effect of garlic oil on acute liver injury induced by acetaminophen (AP) in mice. Methods 75 male KM mice were divided into 5 groups: negative control, model and three levels of garlic oil groups (25, 50, 100 mg/kg). After pretreated with garlic oil 2 h, model group and garlic oil groups were lavaged AP (240 mg/kg). After 24 hours, blood sampling from the eye ball was performed to test the activities of alanine transaminase (ALT) and aspartate amino transferase (AST). Meanwhile, the changes of liver index and hepatic histopathology were determined. Results Compared with the model group, the liver index and the activities of ALT and AST of garlic oil groups were significantly decreased(P<0.01). Ballooning degeneration, even necrosis of hepatocytes and infiltration of a number of lymphocytes were observed in model group, while the garlic oil groups had not distinct pathologic changes. Conclusions Garlic oil can effectively protect mice against acute liver injury induced by AP.
